摘要
目的探讨管道瓦斯爆炸致大鼠烧冲复合伤的特点和建模条件。方法取116只Sprague Dawley大鼠分为实验组(n=96)和对照组(n=20)。实验组大鼠在φ2000瓦斯爆炸系统进行瓦斯爆炸,对照组大鼠不进行瓦斯爆炸,其他实验条件同实验组。爆炸结束后,观察实验组大鼠一般情况及烧冲复合伤发生情况。爆炸实验结束后24 h,取2组存活大鼠,用体积分数1%戊巴比妥钠(40 mg·kg^(-1))麻醉后处死,取支气管和肺组织;观察支气管和肺大体形态,判断肺损伤程度。取2组大鼠石蜡包埋支气管及肺组织,行苏木精-伊红染色,光学显微镜下观察大鼠肺和支气管组织病理形态;应用免疫组织化学法检测支气管及肺组织中caspase-3及caspase-9的表达。结果实验组大鼠体表烧伤发生率为100.0%,69.7%为体毛烧伤、部分区域结痂,累及皮肤烧伤共7例,均为Ⅱ度烧伤,分布于头及肢体末端、腹部、躯干等部位;30.3%为体毛轻度烧伤。存活大鼠表现为精神差、趴卧不起、拒食、呼吸加速。实验组大鼠烧冲复合伤及肺损伤发生率均为100.0%;大鼠现场共死亡19例,死亡率为19.8%,实验结束即时死亡17例,2 h内死亡2例。对照组大鼠支气管及肺组织结构正常,肺泡结构及支气管黏膜和黏膜肌内组织完整。实验组大鼠支气管及肺组织颜色加深,支气管黏膜可见散在出血灶,肺组织表面呈点状及片状出血灶;肺泡间隔明显增宽,血管充血、灶状出血,肺泡腔内见淡粉色水肿液;支气管黏膜上皮细胞部分脱落,管壁充血、水肿,中性粒细胞及淋巴细胞浸润,黏膜肌内见大量中性粒细胞及淋巴细胞浸润,黏膜腺体腺腔内分泌物增多。2组大鼠肺泡上皮细胞、肺间质以及支气管黏膜上皮细胞中均有caspase-3及caspase-9表达。实验组大鼠支气管组织中caspase-3及caspase-9阳性表达率显著高于对照组(χ^(2)=68.310、62.986,P<0.05);实验组大鼠肺组织中caspase-3及caspase-9�
Objective To investigate the characteristics and modeling conditions of combined burn-blast injury in rats caused by pipeline gas explosion.Methods A total of 116 Sprague Dawley rats were divided into experimental group(n=96)and control group(n=20).Rats in the experimental group were performed with gas explosion inφ2000 gas explosion system,the rats in the control group did not carry out gas explosion,and the other experimental conditions were same as those in the experimental group.After the explosion,the general situation and the occurrence of combined burn-blast injury of rats in the experimental group were observed.At 24 hours after explosion,the surviving rats in the two groups were anesthetized with 1%pentobarbital sodium(40 mg·kg^(-1))and then were killed,and the bronchi and lung tissues were taken;the general morphology of bronchus and lung were observed and the degree of lung injury was judged.The bronchi and lung tissues of rats in the two groups were embedded in paraffin,stained with hematoxylin-eosin staining,and the pathological morphology of lung and bronchus was observed under optical microscope;the expressions of caspase-3 and caspase-9 in the bronchi and lung tissues were detected by immunohistochemistry.Results The incidence of body surface burns of rats in the experimental group was 100.0%,among them,69.7%of body surface burns were body hair burns,scabs in some areas,and 7 rats hadⅡdegree skin burns,which was distributed in the ends of head and limbs,abdomen,trunk and other parts;30.3%of body surface burns were slightly burned.The surviving rats in the experimental group showed poor spirit,lying down,refusal to eat and accelerated breathing.The incidence of combined burn-blast injury and lung injury of rats in the experimental group was all of 100.0%.A total of 19 rats died on site,with a mortality rate of 19.8%;17 rats died immediately after the end of the experiment and 2 rats died within 2 hours.In the control group,the structure of bronchi and lung of rats was normal,and the alveolar st
