肥胖症脂肪组织中环状RNA表达水平及功能研究

Dysregulated expression profiles of circular RNA in visceral adipose tissue of obesity individuals

摘要 目的分析肥胖症内脏脂肪组织中环状RNA(circRNA)表达水平。方法留取北京朝阳医院2020年1月至9月收治患者的内脏脂肪组织,根据体重指数,分肥胖(>32.5 kg/m^(2))和正常组(<20.0 kg/m^(2)),选取10份(肥胖组6份,正常组4份)行全转录组测序,比较circRNA表达水平并利用生物信息学方法分析其功能。以Log2(Fold change>1)且P<0.05筛选差异化表达的circRNA,以P<0.05为条件筛选富集的通路和分子功能。结果与正常组比较,肥胖组有52个差异表达的circRNA[Log2(Fold change)>1,且P<0.05],其宿主基因主要富集在"胰岛素抵抗"、"肿瘤坏死因子通路"和"细胞因子-因子受体通路"等通路(P<0.05),筛选出上述通路中的关键基因,如趋化因子(CX3CL1)、丝裂原活化蛋白激酶(MAPK)、白细胞介素6(IL-6)、胰岛素受体底物2(ISR2)等,这些基因与慢性炎性反应密切相关。结论circRNAs可能通过调控慢性炎性反应在肥胖症发生发展中发挥作用。 Objective To compare expression profiles of circular RNA(circRNA)in visceral adipose tissue(VAT)of obesity[body mass index(BMI)>32.5 kg/m^(2)]and lean(BMI<20.0 kg/m^(2))individuals.Methods The whole transcriptome RNA sequencing was performed to detect the expression of circRNAs,and bioinformatic methods were used for function analysis,circRNAs with a Log2(fold change)>1 and P value of<0.05 were considered as dysregulated,and the enriched pathways with a P value of<0.05 were selected for further analysis.Results A total of 52 circRNAs were differentially expressed in VAT between obesity and lean individuals,including 25 up-and 27 down-regulated circRNAs[Log2(fold change)>1,P<0.05].Pathway enrichment analysis indicated that those circRNAs were mainly involved in insulin resistance,TNF pathway,cytokine-cytokine receptor interaction(P<0.05).A further analysis selected hub genes enriched in those Kyoto gene and genome encyclopedia(KEGG)pathways,which were mitogen activated kinase-like protein(MAPK),interleukin 6(IL-6),insulin receptor substrate 2(ISR2),all these genes were associated with chronic inflammation.Conclusion The circRNA might play an important role in the pathogenesis of obesity by regulating inflammation.
