心肌缺血再灌注损伤对围绝经期大鼠认知功能和雌激素水平的影响 被引量:1

摘要 目的探究心肌缺血再灌注损伤对围绝经期大鼠认知功能和雌激素水平的影响。方法60只15~20月龄雌性SD大鼠采用随机数字表法分为3组:对照组(Con组)、假手术组(sham组)和心肌缺血再灌注组(MIR组),每组20只。3组大鼠进行6 d水迷宫定向航行试验后,MIR组采用结扎冠状动脉左前降支30 min再灌注120 min的方法制备心肌缺血再灌注模型,sham组仅穿线不结扎,Con组不予手术处理。MIR组于结扎前和造模后采集血液样本,采用ELISA法测血清cTnI浓度和血清雌激素浓度;sham组和Con组仅于造模后采集血液样本,采用ELISA法测血清雌激素浓度。造模1 d后采用水迷宫空间探索试验评估大鼠运动和认知水平。结果MIR组造模后血清cTnI浓度较缺血前显著升高(P<0.05)。与Con组相比,sham组和MIR组大鼠穿越平台次数显著减少(P<0.05);与sham组相比,MIR组大鼠穿越平台次数显著减少(P<0.05)。3组大鼠造模后雌激素水平差异无统计学意义(P>0.05)。结论心肌缺血再灌注损伤会引发围绝经期大鼠海马依赖性认知功能下降,但不影响围绝经期大鼠血清雌激素水平。 Objective To explore the effect of myocardial ischemia/reperfusion injury on the cognitive function and the level of estrogen in menopause rats.Methods Sixty female SD rats,aged 15-20 months,were randomly divided into three groups(n=20):control group(Con group),sham operation group(sham group)and myocardial ischemia/reperfusion group(MIR group).The rats in three groups were analyzed in the place navigation test for 6 d,and then the MIR model rats in MIR group were induced by 30 min occlusion of anterior descending branch of left coronary artery(LAD)followed by 120 min reperfusion.In sham group,the rats were only threaded around LAD but not ligated.The rats in Con group received no treatment.Blood samples in MIR group were obtained before occlusion and after modeling to determine the concentration of cardiac troponin I(cTnI)and the level of estrogen in serum by ELISA.Blood samples in sham group and Con group were obtained after the modeling to detect the level of estrogen in serum by ELISA.Motor and cognitive functions of rats were analyzed in the space navigation test at day 1 after the modeling.Results Compared with that before ischemia,serum cTnI level was significantly increased after modeling(P<0.05).Compared with Con group,the frequency of crossing the original platform was decreased in sham group and MIR group(P<0.05).Compared with sham group,the frequency of crossing the original platform was decreased in MIR group(P<0.05).The level of estrogen after the modeling had no significant difference between three groups(P>0.05).Conclusion Myocardial ischemia/reperfusion injury induces the cognitive function reduction in the menopause rats,but has no effect on serum estrogen.
