摘要
皖西白鹅耐粗饲、抗病能力强和耗料少,但季节性繁殖,繁殖性能和产蛋量低。为分析不同饲养水平对鹅就巢性的影响,本试验检测了皖西白鹅产蛋期中与就巢相关基因的表达差异。将含有不同硼酸添加量的饲料饲喂皖西白鹅,试验期结束个体屠宰后采集下丘脑组织,提取组织总RNA,反转录后获得cDNA,通过实时荧光定量PCR检测GRB2、HSP70和AMPK基因的表达水平。结果表明,与对照组相比,添加不同浓度硼都会使GRB2、HSP70和AMPK基因的表达极显著下调(P<0.01)。综上,饲料中添加硼可使下丘脑中的GRB2、HSP70和AMPK基因表达下调,进一步从分子水平揭示皖西白鹅产蛋性能的潜在调控机制。
The Wanxi white geese were tolerant to rough feeding,resistant to disease and consumes less material,but breeds seasonally and has low reproductive performance and egg production.The differences in the expression of broody-related genes were examined during the egg-laying period in Wanxi white geese,to analyze the effects of different feeding levels on the expression of these genes through this experiment.The hypothalamus tissues were collected at the end of the test period after individual slaughter and total RNA was extracted.The cDNA was obtained by reverse transcription and the expression levels of GRB2,HSP70 and AMPK genes were measured by real-time fluorescence quantitative PCR.The results showed that the addition of boron at different concentrations caused a highly significant down-regulation of the expression of GRB2,HSP70 and AMPK genes compared to the control group(P<0.01).Overall,the addition of boron to the diet resulted in the down-regulation of GRB2,HSP70 and AMPK gene expression in the hypothalamus,which is to further reveal the potential regulation mechanism of egg-laying performance of Wanxi white geese at the molecular level.
