利用转录组测序分析GRB2和SOS基因在籽鹅等级前卵泡中的mRNA表达 被引量:3

摘要 为研究生长因子受体结合蛋白2(Growth factor receptor-bound protein 2,GRB2)和SOS(Son of sevenless)基因组在籽鹅等级前卵泡中的表达情况,本研究采用高通量测序技术对籽鹅等级前卵泡进行转录组测序分析,之后通过荧光定量PCR进行验证。通过FPKM值进行分析,GRB2和SOS在籽鹅等级前卵泡中均有表达。随着卵泡的发育,GRB2在小白卵泡中表达量最低,在初级卵泡中表达量最高;SOS在大白卵泡和中白卵泡中的表达量最低,在初级卵泡中的表达量最高。荧光定量PCR结果与转录组测序结果基本一致。 The expression of growth factor receptor-bound protein 2 (GRB2) and Son of sevenless (SOS) in the pre-follicles of Zi goose was studied by transcriptome sequencing technology. The Zi-goose pre-hierarchical follicles transcriptome was sequencing with the Illumina Hiseq 2500 and the result was indicated by fluorescence quantitative PCR. The results showed that GRB2 and SOS gene all were expressed in pre-hierarchieal follicles though computational analysis the FPKM value. With the development of follicles,the expression of GRB2 had the minimum expression level in the middle white follicles, and the primary follicles had the highest expression. The expression of SOS had the minimum expression level in the large white follicles and the middle white follicels, and the primary follicles had the highest expresssion. The transcriptome results were basically consistent with fluorescent quantitative PCR.
