摘要
目的评价淫羊藿破壁饮片质量。方法淫羊藿饮片经粉碎,过80目筛,得传统饮片;再经超微粉碎成超微粉(D90<45μm),得破壁粉;经成型工艺,得破壁饮片。建立12批淫羊藿传统饮片、破壁粉、破壁饮片的高效液相色谱(HPLC)指纹图谱,计算指标性成分朝藿定A、朝藿定B、朝藿定C、淫羊藿苷、宝藿苷Ⅰ的含量,色谱柱采用OdyssiL C18柱(250 mm×4.6 mm,5μm),流动相为水-乙腈(梯度洗脱),流速为1.0 mL/min,检测波长为270 nm,柱温为30℃,进样量为10μL。结果朝藿定A、朝藿定B、朝藿定C、淫羊藿苷、宝藿苷Ⅰ质量浓度分别在0.04~1.84 mg/mL、0.03~1.69 mg/mL、0.03~1.81 mg/mL、0.02~0.15 mg/mL、0.03~1.99 mg/mL范围内与峰面积线性关系良好(r=0.9990,n=7);平均加样回收率分别为101.01%,103.84%,99.25%,103.40%,102.65%,RSD分别为1.37%,0.95%,0.74%,0.81%,0.31%(n=9);精密度和重复性试验结果的RSD均小于2.0%(n=6)。传统饮片、破壁粉与破壁饮片共标定13个共有峰;共有模式相关性分析显示,传统饮片、破壁粉及破壁饮片与对照图谱(R)的相似度分别为0.996,0.998,0.998,相似度均大于0.990;5个黄酮类指标性成分总含量从高到低依次为破壁粉>破壁饮片>传统饮片,质量分数分别为6.43%,5.76%,5.43%。结论在相同提取工艺下,淫羊藿经破壁粉碎、成型后可提高黄酮类成分含量,同时保证破壁前后质量的稳定性。
Objective To evaluate the quality of Epimedii Folium cell-wall broken decoction pieces.Methods Epimedii Folium decoction pieces were crushed and passed through an 80-mesh sieve to obtain the traditional decoction pieces.Then they were crushed into ultrafine powder(D90<45μm)to obtain the cell-wall broken powder.Through the molding process,the cell-wall broken decoction pieces were obtained.The high-performance liquid chromatography(HPLC)fingerprints of 12 batches of Epimedii Folium traditional decoction pieces,cell-wall broken powder and cell-wall broken decoction pieces were established,and the content of index components epimedin A,epimedin B,epimedin C,icaritin and baohuosideⅠwas calculated.The chromatographic column was OdyssiL C18 column(250 mm×4.6 mm,5μm),the mobile phase was water-acetonitrile(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 270 nm,the column temperature was 30℃and the injection volumn was 10μL.Results The linear ranges of epimedin A,epimedin B,epimedin C,icaritin and baohuosideⅠwere 0.04-1.84 mg/mL,0.03-1.69 mg/mL,0.03-1.81 mg/mL,0.02-0.15 mg/mL,0.03-1.99 mg/mL(r=0.9990,n=7).The average recoveries of epimedin A,epimedin B,epimedin C,icaritin and baohuosideⅠwere 101.01%,103.84%,99.25%,103.40%and 102.65%with RSDs of 1.37%,0.95%,0.74%,0.81%and 0.31%(n=9).RSDs of precision and repeatability tests were less than 2.0%(n=6).Thirteen common peaks were obtained for traditional decoction pieces,cell-wall broken powder and cell-wall broken decoction pieces.The correlation analysis of common patterns showed that the similarities of traditional decoction pieces,cell-wall broken powder and cell-wall broken decoction pieces with the reference chromatograms(R)were 0.996,0.998 and 0.998,respectively,all of which were greater than 0.990.The total content of five flavonoid index components was the highest in the cell-wall broken powder,followed by the cell-wall broken decoction pieces and traditional decoction pieces,and the mass fractions were 6.43%,5.76%and 5.43%,resp
