摘要
为探讨分离自患病刺参(Apostichopus japonicus)的一株溶藻弧菌(Vibrio alginolyticus)感染刺参后,刺参体腔细胞免疫相关基因的响应变化,分别给刺参注射100μL生理盐水(对照组)和1×10^(9) CFU/mL溶藻弧菌菌液(实验组),用荧光定量PCR技术,检测24 h内刺参体腔细胞Lys、Rel和p105基因表达的变化。结果表明,注射生理盐水对刺参体腔细胞Lys、Rel和p105基因表达量无显著影响。感染溶藻弧菌后,实验组刺参体腔细胞中溶菌酶基因的表达量在24 h显著升高,且1 h和24 h时的表达量显著高于对照组(P<0.05);刺参体腔细胞中p105基因在感染后表达量呈上升趋势,3 h达到峰值,在3 h和6 h,p105基因的表达量显著高于对照组(P<0.05);刺参体腔细胞Rel基因表达量在感染后无显著变化。可见刺参感染溶藻弧菌后其体腔细胞Lys和p105基因的表达在短时间内迅速响应。
In order to explore the response of immune-related genes including Lys,Rel and p105 in coelomocytes of sea cucumber Apostichopus japonicus to the infection of Vibrio alginolyticus(isolated from diseased sea cucumber),sea cucumbers were respectively injected with 100μL normal saline(control group)and V.alginolyticus at 1×10^(9) CFU/mL(experimental group).The results showed that injection of normal saline had no significant effect on the expression of Lys,Rel and p105 genes in coelomocytes of A.japonicus.After infection with V.alginolyticus,the expression of Lys gene in the coelomocytes of A.japonicus increased significantly at 24 h,and the expression of Lys gene in the experimental group at 1 h and 24 h was significantly higher than those in the control group(P<0.05).The expression of p105 gene in A.japonicus coelomocytes increased after infection,reached the peak at 3 h,and the expression of p105 gene at 3 h and 6 h was significantly higher than those in the control group(P<0.05).The expression of Rel gene in coelomocytes of sea cucumber had no significant change after infection with V.alginolyticus.It can be concluded that the expression of Lys and p105 genes in coelomocytes of A.japonicus infected with Vibrio alginolyticus responded rapidly.
