摘要
为给红锥(Castanopsis hystirx)人工授粉和新品种培育提供基础,测定红锥花粉萌发率和花粉管长度,探讨花粉的最佳离体萌发条件和贮藏条件。结果表明,采用100 g/L蔗糖+200 mg/L硼酸培养液,培养温度为25℃,花粉萌发率最高(28.23%),花粉管长度最长(260.45μm);采用-20℃+干燥剂或-80℃+干燥剂贮藏花粉效果较好,贮藏365天,萌发率仍有30.54%和26.43%,花粉管长度为108.61和95.65μm。低温、干燥有利于红锥花粉贮藏。
In order to provide a basis for artificial pollination and new variety breeding of Castanopsis hystrix,Germination rate and pollen tube length of C.hystrix pollen were measured to explore optimal condition for pollen germination in vitro and storage condition.Results showed that the highest pollen germination rate(28.23%)and the longest pollen tube length(260.45μm)were achieved by using 100 g/L sucrose+200 mg/L boric acid culture solution and incubation temperature of 25℃.Pollen storage at-20℃+desiccant or-80℃+desiccant was more effective,with 30.54%and 26.43%germination rates and 108.61 and 95.65μm pollen tube lengths after 365 d.Low temperature and dryness were favorable for pollen storage of C.hystrix.
