小儿柴胡退热栓的解热作用研究 被引量:1

摘要 目的评价小儿柴胡退热栓的解热作用,并初步探讨其解热机制。方法将新西兰家兔25只随机分为空白组、模型组、试药组(小儿柴胡退热栓)和阳性对照组(柴胡注射液组、小儿布洛芬栓组),通过建立家兔发热模型,观测各组家兔行为学和肛温变化,ELISA检测血浆中白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平和下丘脑中环磷酸腺苷(cAMP)、精氨加压素(AVP)和前列腺素E_(2)(PGE_(2))水平。结果与模型组相比,小儿柴胡退热栓在给药后1~2.5 h内可显著降低发热家兔肛温至正常水平(P<0.01);同时小儿柴胡退热栓能显著下调发热家兔血浆中IL-1β、IL-6、TNF-α水平(P<0.05);并可显著下调发热家兔下丘脑中cAMP(P<0.05)、PGE_(2)(P<0.01)水平,上调AVP水平(P<0.01)。结论小儿柴胡退热栓可显著降低发热家兔体温,其机制可能与调节发热家兔血浆中IL-1β、IL-6、TNF-α含量及下丘脑中cAMP、PGE_(2)、AVP含量密切相关。 Objective To evaluate the antipyretic effect of Xiaoer Chaihu Tuire suppository,and determine the mechanism.Methods Totally 25 New Zealand rabbits were randomly divided into a blank groups,a model group,a drug group(Xiaoer Chaihu Tuire suppository),and two positive control groups(Chaihu injection group and paediatric ibuprofen suppositories group).The endotoxin-reduced rabbit fever model was used,and the changes of behaviors and rectal temperature in every group were observed.The levels of IL-1β,IL-6,and TNF-αin the plasma and cAMP,AVP,and PGE_(2) in the hypothalamus were measured by ELISA.Results During 1~2.5 hours after the administration,Xiaoer Chaihu Tuire suppository significantly reduced the rectal temperature as compared with the model group(P<0.01).Xiaoer Chaihu Tuire suppository significantly reduced the level of IL-1β,IL-6,and TNF-αin the plasma(P<0.05),reduced the level of cAMP(P<0.05)and PGE_(2)(P<0.01),and raised the level of AVP(P<0.01)in the hypothalamus.Conclusion Xiaoer Chaihu Tuire suppository have antipyretic effect on rabbits with fever,whose mechanism is closely related to the regulation of the levels of IL-1β,IL-6,and TNF-αin the plasma and cAMP,PGE_(2),and AVP in the hypothalamus.
