摘要
目的探究Wnt信号通路中ZNRF3和RNF43对儿童急性B淋巴细胞白血病(B-ALL)的意义。方法收集41例B-ALL患儿初诊(ND组)及化疗第33天的骨髓样本。初诊时依据危险度分为低危组(n=13)、中危组(n=17)和高危组(n=11)。41例患儿于化疗第33天骨髓均达到完全缓解(CR),定为CR组。采用实时荧光定量聚合酶链反应(qRT-PCR)检测ND组、CR组ZNRF3及RNF43 mRNA表达,采用蛋白印迹法(Western blot)检测其蛋白表达。结果ND组ZNRF3、RNF43的mRNA及蛋白表达较CR组明显减低(P<0.05)。低危组ZNRF3、RNF43的mRNA及蛋白表达明显高于中、高危组(P<0.05)。结论初诊时ZNRF3和RNF43低表达可能与儿童ALL发生相关,其相对高表达可能与低危险度相关。
Objective To investigate the significance of zinc and ring finger 3(ZNRF3)and ring finger protein 43(RNF43)in the pathogenesis of pediatric B-cell acute lymphoblastic leukemia(B-ALL).Methods A total of 41 newly diagnosed B-ALL children who had finished induction therapy were enrolled.The children pre-treatment were included in group of newly diagnosed,and they were included in group of remission(CR)after 33-days induction therapy when they all achieved complete remission.According to the risk-stratification,the ND group was divided into low risk(n=13),intermediate risk(n=7)and high risk(n=11)groups.Two milliliter bone marrow samples were collected from all patients above.QRT-PCR was used to determine the mRNA expression of ZNRF3 and RNF43 in bone marrow mononuclear cells.Western blot was used to detect the protein expression of ZNRF3 and RNF43.Results The mRNA and protein expression of ZNRF3 and RNF43 in ND group was significantly lower than that in CR group(P<0.05).The mRNA and protein expression of ZNRF3 and RNF43 in the low-risk group were significantly higher than those in the intermediate-and high-risk groups(P<0.05).Conclusions Low expression of ZNRF3 and RNF43 may be associated with the pathogenesis of pediatric B-ALL,and the increase of their expression may be related to low risk.
