金银花茯苓提取物对小鼠免疫功能的在体调控作用 被引量:4

摘要 目的研究金银花茯苓提取物对免疫抑制小鼠免疫功能的在体调控作用。方法应用水提法制备金银花茯苓提取物。实验小鼠随机分为6组:空白组,模型组,金银花茯苓复配低剂量组、中剂量组、高剂量组,阳性对照组。模型组,金银花茯苓提取物低剂量组、中剂量组、高剂量组,阳性对照组的小鼠连续3 d腹腔注射给予环磷酰胺(cyclophosphamide,CTX)80 mg·kg^(-1);空白对照组给予等体积生理盐水。CTX给药后,连续灌胃小鼠金银花茯苓提取物7 d,其中给药剂量分别为低剂量组(90 mg·kg^(-1))、中剂量组(180 mg·kg^(-1))、高剂量组(360 mg·kg^(-1))。阳性对照组灌胃给予左旋咪唑(10 mg·kg^(-1))。空白对照组与模型对照组连续灌胃7 d等体积的生理盐水。灌胃后取小鼠脾脏、胸腺检测小鼠免疫器官指数,眼眶取血进行小鼠全血分析,制备脾细胞悬液测定ConA诱导淋巴细胞增殖、淋巴细胞分群、NK细胞活性以及脾细胞凋亡。结果金银花茯苓提取物有改善由CTX导致的免疫器官损伤的趋势,提高免疫抑制模型小鼠脾脏NK细胞杀伤能力,提高免疫抑制模型小鼠脾淋巴细胞亚群CD4^(+)/CD8^(+)比值,减少由CTX导致的脾细胞凋亡,增加脾淋巴细胞ConA诱导增殖,降低由CTX引起的白细胞增多,逆转CTX引起的血红蛋白浓度下降。结论金银花茯苓提取物可逆转环磷酰胺所导致的小鼠免疫抑制,提升小鼠的免疫功能,研究结果表明金银花茯苓提取物具有提高免疫的作用,为后续提高免疫力保健食品和药品的研发提供了理论基础。 Objective To study the in vivo regulation of Lonicerae Japonicae Flos and Poria extract on immune function in immunosuppressive mice.Methods The extract of Lonicerae Japonicae Flos and Poria was prepared by water extraction.The experimental mice were randomly divided into 6 groups:control group,model group,Lonicerae Japonicae Flos and Poria complex low dose group,medium dose group,high dose group and positive group.The mice of model group,Lonicerae Japonicae Flos and Poria extract low dose group,medium dose group,high dose group and positive group were intraperitoneally injected with cyclophosphamide(CTX)80 mg·kg^(-1)for 3 days,and the blank control group was given equal volume normal saline.After administration of CTX,mice were continuously gavaged with Lonicerae Japonicae Flos and Poria extract for 7 days.The doses were low dose group(90 mg·kg^(-1)),medium dose group(180 mg·kg^(-1))and high dose group(360 mg·kg^(-1)).The positive group was given levamisole(10 mg·kg^(-1))by gavage.The blank control group and the model control group were gavaged with equal volume of normal saline for 7 days.After intragastric administration,the spleen and thymus of mice were taken to detect the index of immune organs,and the blood from orbit was taken for whole blood analysis.The splenocyte suspension was prepared to determine the lymphocyte proliferation,lymphocyte clustering,NK cell activity and splenocyte apoptosis induced by ConA.Results Lonicerae Japonicae Flos and Poria extract had the tendency to improve the immune organ damage caused by CTX,improve the killing ability of spleen NK cells in immunosuppressive model mice,increase the CD4^(+)/CD8^(+)ratio of spleen lymphocyte subsets in immunosuppressive model mice,reduce the apoptosis of spleen cells caused by CTX,increase the ConA induced proliferation of spleen lymphocytes,and reduce the increase of white blood cell caused by CTX,Reverse the decrease of hemoglobin concentration caused by CTX.Conclusion Lonicerae Japonicae Flos and Poria extract can reverse the immunos
