摘要
LPS为1型T非依赖性抗原,可以在没有其他细胞辅助的情况下直接刺激成熟B淋巴细胞的增殖和分化并产生抗体。为了研究LPS诱导B淋巴细胞增殖和分化的调控机制,本研究检测了一些关键蛋白分子及转录因子表达的改变。LPS诱导的原代B淋巴细胞的分化伴随着B淋巴细胞抗原受体(BCR)信号转导相关分子表达的显著降低,说明BCR的功能在浆细胞阶段已处于次要的地位。我们的结果还证明,LPS可以在原代B淋巴细胞有效地诱导被称为“B淋巴细胞终极分化主调控子”的转录因子Blimp-1的表达,并抑制转录因子Bcl-6和BSAP的表达,这一变化同B淋巴细胞的发育同步,可能在诱导增殖与分化的调控中起着关键作用。
LPS is a type 1 T,independent antigen and can induce proliferation and differentiation of the mature B lymphocytes and the production of antibodies without the help of other cells. To study the modulating mechanisms of LPS,induced proliferation and differentiation of B lymphocytes,the alterations of expression of the key protein molecules involving B cell antigen receptor(BCR) signaling pathways and some transcription factors in LPS,treated mouse primary splenic B lymphocytes were investigated in the present study. The results of this investigation showed that the expression of numerous genes involving in the BCR signaling pathways was down,regulated during the LPS,induced B cell differentiation. It was also found that LPS could induce the expression of transcription factor Blimp,1, the so called'master regulator of the terminal B cell differentiation', and repress the expression of transcription factor Bcl,6 and BSAP. The differential expression of the transcription factors might play an important role in the regulation of the LPS,induced B cell differentiation.
