解脂耶氏酵母合成萜烯的进展:见微知著的改造策略 被引量:3

摘要 萜烯是一类基于五碳单元类异戊二烯的天然化合物,种类繁多且具有多种生物活性,被广泛应用于食品、医药和化工领域。传统萜类物质生产依赖于化学合成或植物组织提取,存在产率低、资源浪费的缺点。近年来,代谢工程和合成生物学的发展促进了微生物细胞工厂的高效构建,为化学品的微生物合成提供了新的选择。解脂耶氏酵母因前体甲羟戊酸途径的内源性、乙酰辅酶A供给的充足性以及底物利用的广谱性,被视为萜类化合物合成的重要底盘生物。本文总结了萜烯合成关键酶的挖掘与改造、代谢途径设计与优化、细胞器水平的区室化以及细胞整体水平调控等解脂耶氏酵母中萜类化合物的合成策略,展望了进一步提高该宿主中萜类化合物产量的研究方向与工程方法。 Terpenes are natural compounds based on C5 unit isoprene, with a variety of biological activities and are widely used in food, pharmaceutical and chemical industries.Traditionally, terpenes production relies on chemical synthesis or plant tissue extraction with low yield.Recently, the development of metabolic engineering and synthetic biology has promoted the efficient construction of microbial cell factories and provided new options for the microbial synthesis of chemicals.Yarrowia lipolytica is regarded as an important chassis for biosynthesis of terpenes due to the inherence of mevalonate pathway, the adequacy of acetyl-CoA supply and the broad-spectrum of substrates utilization.This review aims to summarize the bioengineering strategies of terpenes synthesis in Y.lipolytica,such as mining and modification of key enzymes in terpene synthesis, design and optimization of biosynthesis pathways, subcellular compartmentalization and cellular regulation.We also propose future research directions and engineering methods for further increasing terpenoids production by Y.lipolytica.
