摘要
目的探讨槐花颗粒抗氧化成分提取液的体外抗氧化活性,以及其体外抑菌作用和作用机制。方法采用乙醇水浴加热提取槐花颗粒的抗氧化成分,制备抗氧化成分提取液(质量浓度为2,4,6,8 mg/mL),以维生素C溶液(质量浓度为7 mg/mL)作阳性对照,考察提取液对1,1-二苯基-2-三硝基苯肼(DPPH)、超氧阴离子、过氧化氢的清除能力,以及对Fe^(3+)的还原能力,并评价其体外抗氧化活性。采用琼脂扩散法测定提取液对金黄色葡萄球菌的抑菌圈直径;采用二倍稀释法测定槐花颗粒的最低抑菌浓度(MIC),采用活菌计数法计数菌落,测定最低杀菌浓度(MBC);通过测定提取液作用下细菌细胞膜电导率和细菌培养液中DNA及RNA等大分子的含量,分析其抑菌机制。结果抗氧化成分提取液质量浓度为2~8 mg/mL时,对DPPH自由基、超氧阴离子、过氧化氢均有较好的清除能力,对Fe^(3+)具有较好的还原能力,且清除能力和还原能力随着质量浓度的增加而增强,对Fe^(3+)的还原能力与维生素C溶液接近。抗氧化成分提取液质量浓度为8 mg/mL时,对DPPH自由基、过氧化氢的清除能力比维生素C溶液更强;对超氧阴离子清除力与维生素C溶液相比较弱。抗氧化成分提取液质量浓度为0.8,1.6,3.1 g/mL时,对金黄色葡萄球菌的抑制效果明显,抑菌圈直径分别为5.0,7.0,10.0 mm;对金黄色葡萄球菌的MIC为48.8 mg/mL,MBC为390.0 mg/mL。随着抗氧化成分提取液质量浓度的升高,培养基电导率逐渐升高,且细菌培养液中DNA及RNA等大分子的含量逐渐增加。结论槐花颗粒抗氧化成分提取液具有良好的抗氧化能力,且抗氧化能力随抗氧化成分质量浓度的升高而增强;同时,具有良好的抑菌效果,可能是通过改变细胞膜和细胞壁的通透性来抑制金黄色葡萄球菌。
Objective To investigate the antioxidant activity in vitro of the antioxidant ingredients extract of HuaiHua Granules,as well as its antibacterial effect and mechanism in vitro.Methods The antioxidant ingredients of HuaiHua Granules were extracted by heating in the ethanol-water bath to prepare the antioxidant ingredients extract with mass concentration of 2,4,6,8 mg/mL.Vitamin C solution(mass concentration of 7 mg/mL)was used as a positive control to investigate the scavenging abilities of the extract to 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH),superoxide anion and hydrogen peroxide,as well as the reducing ability to Fe3+,and its antioxidant activity in vitro was evaluated.The agar diffusion method was used to determine the antibacterial circle diameter of the extract against Staphylococcus aureus,the double dilution method was used to determine the minimum inhibitory concentration(MIC)of HuaiHua Granules,the viable count method was used to count the colonies and determine the minimum bactericidal concentration(MBC),the conductivity of the bacterial cell membrane and the contents of DNA,RNA and other macromolecules in the bacterial culture medium under the action of the extract were determined to analyze the antibacterial mechanism.Results When the mass concentration of the antioxidant ingredients extract was 2-8 mg/mL,it had good scavenging abilities for DPPH free radical,superoxide anion and hydrogen peroxide and good reducing ability for Fe^(3+),and the scavenging ability and reducing ability increased with the increase of the mass concentration.The reducing ability of the extract to Fe^(3+)was close to that of the vitamin C solution.When the mass concentration of the antioxidant ingredients extract was 8 mg/mL,the scavenging abilities of the extract to DPPH free radical and hydrogen peroxide were stronger than that of the vitamin C solution,and the scavenging ability of the extract to superoxide anion was weaker than that of vitamin C solution.When the mass concentration of the antioxidant ingredients ex
