摘要
目的:考察^(60)Co-γ射线辐照灭菌对赶黄草中6种成分(原儿茶酸、没食子酸、槲皮素、芦丁、芹菜素和山柰酚)含量的影响。方法:分别选择剂量为0,2,5,8 kGy的;Co-γ射线对赶黄草药材进行辐照。采用Waters Sunfire ODS C_(18)色谱柱(250 mm×4.6 mm, 5μm),流动相为A-乙腈,B-0.05%磷酸水溶液系统,梯度洗脱,检测波长340 nm,流速为1.0 ml·min^(-1),柱温35℃,比较辐照前后活性成分含量变化,采用偏最小二乘-判别分析(PLS-DA)对其总体质量进行分析评价。结果:赶黄草药材中原儿茶酸、没食子酸、槲皮素、芦丁、芹菜素和山柰酚分别在0.069~1.727 mg·ml^(-1)、0.012~0.311 mg·ml^(-1)、0.038~0.945 mg·ml^(-1)、0.031~0.780 mg·ml^(-1)、0.005~0.126 mg·ml^(-1)、0.008~0.208 mg·ml^(-1)范围内呈良好线性,平均加样回收率分别为99.9%、99.5%、99.9%、99.5%、98.6%和97.2%。辐照剂量不超过5 kGy时,各组分含量差异无统计学意义(P>0.05),PLS-DA可将不同辐照处理的样品显著分类,并筛选出导致质量差异的5个差异标志物。结论:为保证药品安全有效,采用;Co-γ射线对赶黄草药材进行灭菌时,辐照剂量不宜超过5 kGy。
Objective: To study the effects of ^(60)Co-γ ray irradiation on the contents of protocatechuic acid, gallic acid, quercetin, rutin, apigenin and kaempferol in Penthorum chinense Pursh. Methods: A Waters Sunfire ODS C_(18)column(250 mm×4.6 mm, 5 μm) was adopted and the wavelength of UV detection was 340 nm at the flow rate of 1.0 ml·min;. The mobile phase consisted of acetonitrile(A) and 0.05% phosphoric acid(B) with gradient elution. The column temperature was set at 35℃. The irradiation doses were 0, 2, 5 and 8 kGy, respectively. The contents of the active ingredients were compared before and after irradiation, and the further quality assessment of the herb was carried out by discriminant analysis of partial least squares(PLS-DA). Results: The linear range of protocatechuic acid, gallic acid, quercetin, rutin, apigenin and kaempferol was 0.069-1.727, 0.012-0.311, 0.038-0.945, 0.031-0.780, 0.005-0.126 mg·ml;and 0.008-0.208 mg·ml^(-1), respectively. The average recoveries were 99.9%, 99.5%, 99.9%, 99.5%, 98.6% and 97.2%, respectively. When the radiation dose was not more than 5 kGy, the content differences of five components had no statistical significance(P>0.05), and PLS-DA screened out 5 differential markers leading to the quality differences by radiant dose. Conclusion: To ensure the safety and efficacy of Penthorum chinense Pursh., the irradiation dose of;Co-γ should be not more than 5 kGy.
