电喷雾串联质谱法鉴定甜叶菊干叶中甜菊糖苷的结构 被引量:4

摘要 为了对甜叶菊中甜菊糖苷类成分的结构进行鉴定。建立了一种利用电喷雾质谱法鉴定甜叶菊中甜菊糖苷类结构的方法,与液质联用技术不同,该方法不需要复杂的前处理过程,能够直接判断样品中含有的多种甜菊糖苷。结果表明,对甜叶菊中的甜菊糖苷进行电喷雾质谱研究,正离子模式下电离成加氢、钠、钾离子,负离子模式下电离成减氢、加氯离子,并在负离子模式进行了串联质谱研究,几种甜菊糖苷失去C13和19位取代基,最终形成m/z318的骨架结构。结论:甜菊糖苷类成分的骨架结构为m/z318,根据正负离子模式下特征电离规律从复杂甜叶菊成分中判断了几种甜菊糖苷,根据串联质谱规律确定其取代基为葡萄糖、鼠李糖,并确定其结构。 To identify the structure of steviol glycosides-like components in stevia.A method was developed to identify the structure of steviol glycosides in stevia using electrospray ionization mass spectrometry.Unlike the liquid mass spectrometry technique,this method does not require a complicated pre-treatment process and directly determine the multiple steviol glycosides contained in the sample.The steviol glycosides in stevia were studied by electrospray mass spectrometry,ionized into hydrogen-added,sodium and potassium ions in positive ion mode,and ionized into hydrogen-minus and chlorine-added ions in negative ion mode,and studied by tandem mass spectrometry in negative ion mode:the loss of C13 and 19-position substituents and the final formation of the skeletal structure of m/z318.Conclusion:The skeleton structure of steviol glycosides components was m/z318,and several steviol glycosides were judged from complex stevia components according to the characteristic ionization law in positive and negative ion modes,and their substituents were identified as glucose and rhamnose according to the tandem mass spectrometry law,and their structures were determined.
