摘要 目的 探讨白细胞介素18 (IL-18)经toll样受体4 (TLR4)/髓样细胞分化因子88 (MyD88)/核转录因子-κB(NF-κB)通路促进支气管平滑肌细胞(BSMC)增殖与迁移。方法 采用不同浓度IL-18、不同时间处理BSMC,确定实验条件。细胞计数试剂盒(CCK-8)测定细胞活性,Transwell小室检测细胞迁移,Western blot法检测相关蛋白的表达。抑制剂Eritoran特异性阻断TLR4/MyD88/NF-κB通路,检测TLR4/MyD88/NF-κB通路阻断后对细胞周期、细胞增殖、细胞迁移及TLR4/MyD88/NF-κB通路相关蛋白的表达的影响。结果 IL-18可促进支气管平滑肌细胞细胞增殖,提高细胞迁移能力,上调TLR4、MyD88和NF-κB p65蛋白水平(P<0.05)。Eritoran可抑制IL-18引起的BSMC细胞增殖,上调G1期细胞百分比,下调S期、G2期细胞百分比,抑制细胞迁移,降低TLR4、MyD88和NF-κB p65蛋白水平(P<0.05)。结论 IL-18可能通过上调TLR4/MyD88/NF-κB通路中信号分子的表达,促进了平滑肌细胞增殖和迁移。 【Objective】 To explore the effect of interleukin-18(IL-18) on the proliferation and migration of bronchial smooth muscle cells through toll-like receptor 4(TLR4)/myeloid differentiation factor 88(MyD88)/nuclear transcription factor-κB(NF-κB)pathway. 【Methods】 Bronchial smooth muscle cell(BSMC) was treated with different concentrations of IL-18 at different time to determine the experimental conditions. Cell counting kit(CCK-8) was used to measure cell activity, cell migration was detected by transwell chamber, and the expression of related proteins was detected by western blot. Eritoran, an inhibitor, specifically blocked TLR4/MyD88/NF-κB pathway, and detected the effects of blocked TLR4/MyD88/NF-κB pathway on cell cycle, cell proliferation,cell migration and expression of TLR4/MyD88/NF-κB pathway-related proteins. 【Results】 IL-18 could promote the proliferation and migration of bronchial smooth muscle cells, and up-regulate the protein levels of TLR4, MyD88 and NF-κB p65(P<0.05).Eritoran could inhibit the proliferation of BSMC induced by IL-18, up-regulate the percentage of cells in G1 phase, down-regulate the percentage of cells in S phase and G2 phase, inhibit cell migration and reduce the protein levels of TLR4, MyD88 and NF-κB p65(P<0.05). 【Conclusion】 IL-18 may promote the proliferation and migration of smooth muscle cells by up-regulating the expression of signal molecules in TLR4/MyD88/NF-κB pathway.
