红景天苷对大鼠肾移植后缺血再灌注损伤的保护作用 被引量:1

摘要 目的观察红景天苷(SDS)对大鼠肾移植后缺血再灌注损伤(IRI)的保护作用。方法70只大鼠随机分为假手术组(Sham组)、模型组(Model组)、SDS组、SDS联合佛波酯(PMA)组[核转录因子-κB(NF-κB)信号通路激活剂]。Model组、SDS组、SDS联合PMA组行左肾移植术及右肾切除术,Sham组行右肾切除术。SDS组、SDS联合PMA组术前3 d分别腹腔注射5 mL SDS(含SDS 400 mg/kg)、5 mL SDS(含SDS 400 mg/kg)+PMA(含PMA 5 mg/kg),Sham组、Model组腹腔注射等体积生理盐水,1次/d,持续3 d。检测外周血辅助性T细胞17(Th17)/调节性T细胞(Treg);检测NF-κB p65、p-NF-κB p65蛋白表达。结果与Model组比较,SDS组Th17/Treg值(0.55±0.06 vs.1.27±0.14)、p-NF-κB p65/NF-κB p65(0.36±0.05 vs.0.95±0.11)降低(P<0.05);与SDS组比较,SDS联合PMA组Th17/Treg值(0.84±0.09 vs.0.55±0.06)、p-NF-κB p65/NF-κB p65(0.80±0.09 vs.0.36±0.05)升高(P<0.05)。结论SDS预处理可保护肾移植后IRI大鼠肾功能,减轻炎症反应,维持免疫平衡,可能通过抑制NF-κB信号通路来实现。 Objective To observe the protective effect of salidroside(SDS)on ischemia-reperfusion injury(IRI)after renal transplantation in rats.Methods A total of 70 rats were randomly divided into Sham group,Model group,SDS group,SDS plus PMA group[nuclear transcription factor-κB(NF-κB)signal pathway activator].Rats in the Model group,SDS group,SDS plus PMA group underwent left kidney transplantation and right kidney nephrectomy,while rats in the Sham group underwent right kidney nephrectomy.The SDS group and SDS plus PMA group were intraperitoneally injected with 5 mL SDS(containing 400 mg/kg SDS),and 5 mL SDS(containing 400 mg/kg SDS)+PMA(containing 5 mg/kg PMA)respectively before operation,while the Sham group and Model group were injected with an equal volume of normal saline before operation,once a day for 3 days.The percentage of peripheral blood helper T cell 17(Th17)/regulatory T cell(Treg)12 hours after operation,and protein expressions of NF-κB p65 and p-NF-κB p65 were detected.Results Compared with the Model group,the SDS group had decreased Th17/Treg(0.55±0.06 vs.1.27±0.14),and p-NF-κB p65/NF-κB p65(0.36±0.05 vs.0.95±0.11)(P<0.05).Compared with the SDS group,the SDS plus PMA group had increased Th17/Treg(0.84±0.09 vs.0.55±0.06),and p-NF-κB p65/NF-κB p65(0.80±0.09 vs.0.36±0.05)(P<0.05).Conclusion SDS pretreatment can protect the renal function of rats after kidney transplantation,reduce inflammation,and maintain immune balance by inhibiting NF-κB signaling pathway.
