摘要
新疆某羊场的绵羊出现发热咳嗽、精神萎靡、食欲减退、嗜睡和呼吸困难等症状。经剖检和细菌分离鉴定、PCR鉴定、药敏实验和小鼠致病性试验等方法,分离到了3株革兰氏阴性球杆菌,根据形状和大小判断疑似多杀性巴氏杆菌和溶血性曼氏杆菌;PCR鉴定方法有效扩增出1100 bp、875 bp和900 bp,3条目的条带,经测序与GenBank数据库中同源序列比对,3株菌分别为多杀性巴氏杆菌A型、F型和溶血性曼氏杆菌A2型;药敏试验显示,3株细菌的混合液对左旋氧氟沙星、乳酸环丙沙星敏感;小鼠致病性试验中,小鼠在18 h内陆续死亡,表明巴氏杆菌和曼氏杆菌毒力强。诊断结果为羊巴氏杆菌及曼氏杆菌感染的诊断和治疗提供参考。
Sheep in a farm in Xinjiang showed symptoms such as fever, cough, lassitude, loss of appetite,lethargy and difficulty in breathing. After autopsy, bacterial isolation and identification, PCR identification,drug sensitivity test and pathogenicity test of mice, three strains of Gram-negative bacillus were isolated, and suspected Pasteurella multocida and Mannheimia haemolytica were identified according to their morphology and size. Three target bands with lengths of 1 100 bp, 875 bp and 900 bp were effectively amplified by PCR.After sequencing and comparison with homologous sequences in GenBank database, the three strains were identified as Pasteurella multocida type A, F and Mannheimia haemolytica type A2 respectively. Drug sensitivity test showed that the mixture of these three strains of bacteria was sensitive to levofloxacin and ciprofloxacin lactate. In the pathogenicity test, the mice died successively within 18 h, indicating the strong virulence of Pasteurella and Mannheimia. The results provided reference for the diagnosis and treatment of Pasteurella and Mannerella infection in sheep.
