摘要
目的采用HPLC法对红芪药材、饮片、标准汤剂与配方颗粒的指纹图谱进行相关性研究,并考察四者之间化学成分的差异。方法采用Wondasil C18-WR色谱柱(4.6 mm×150 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,流速1.0 mL·min-1,检测波长280 nm,柱温35℃,建立15批红芪药材、饮片、标准汤剂与配方颗粒的指纹图谱。结果15批红芪饮片中毛蕊异黄酮葡萄糖苷、芒柄花苷、毛蕊异黄酮、7-羟基-4'-甲氧基异黄酮的平均含量略低于药材,配方颗粒中4种有效成分的平均含量略低于标准汤剂;15批红芪药材、饮片、标准汤剂与配方颗粒的HPLC指纹图谱确认了13个共有特征峰,并指认出毛蕊异黄酮葡萄糖苷、芒柄花苷、毛蕊异黄酮、7-羟基-4'-甲氧基异黄酮4种化学成分。结论红芪药材、饮片、标准汤剂与配方颗粒的主要化学成分基本相同;HPLC指纹图谱反映了四者多成分的整体面貌,为红芪配方颗粒的质量控制提供了参考。
Objective To determine the correlation of fingerprint of raw herbs,decoction pieces,standard decoction,and dispensing granules of Hedysari Radix by HPLC,and observe the difference of the main chemical components in different forms.Methods Wondasil C18-WR column(4.6 mm×150 mm,5μm)was used with mobile phase consisting of acetonitrile and 0.1% phosphoric acid aqueous solution at 1.0 mL·min-1,with detection wave of 280 nm,and temperature of 35℃.The HPLC fingerprint of 15 batches of raw herbs,decoction pieces,standard decoction and dispensing granules of Hedysari Radix were established.Results The average content of calycosin-7-glucoside,ononin,calycosin and 7-hydroxy-4’-methoxy isoflavone in the 15 batches of decoction pieces were slightly lower than that of the raw herbs,and the average contents of the 4 active ingredients in the dispensing granules were slightly lower than that of the standard decoction.There were 13 common peaks in all the fingerprint of the 15 batches of raw herbs,decoction pieces,standard decoction and dispensing granules of Hedysari Radix,presenting a good correlation.Four out of the 13 common peaks were indentified calycosin-7-glucoside,ononin,calycosin,and 7-hydroxy-4’-methoxy isoflavone.Conclusion The main chemical components of raw herbs,decoction pieces,standard decoction and dispensing granules of Hedysari Radix are basically the same.The HPLC fingerprint reflects the overall situation of components in different forms.It can provide the reference for the quality control of dispensing granules of Hedysari Radix.
