藏羊源沙克乳酸杆菌的分离、鉴定及体外益生特性研究 被引量:1

摘要 研究旨在获得可用于微生态制剂的优良乳酸杆菌,无菌采集藏羊胃肠道内容物,通过MRS琼脂培养基选择分离,利用细菌形态、生理生化特性、16S rDNA基因序列同源分析等方法进行菌株鉴定,通过抑菌试验、耐受性试验、生长曲线产酸能力试验和药敏试验等对分离菌株的益生特性进行鉴定。结果显示,分离菌株可发酵纤维二糖、麦芽糖、蔗糖和乳糖,16S rDNA序列与参考沙克乳酸杆菌同源性达99%以上。分离菌株对大肠杆菌、沙门氏菌、金黄色葡萄球菌、产气荚膜梭菌等菌株均具有一定的抑制作用,对高温有较强耐受性,对酸性和碱性环境有较强抵抗力,产酸能力强,对新霉素等11种药物敏感。研究表明,试验所得沙克乳酸杆菌对酸碱、高温环境均具有较强耐受性,抑菌能力较强,生长速度和产酸能力也较强,可以为制备藏羊源微生态制剂提供良好的基础菌株。 The study was to obtain good Lactobacillus that could be used in probiotics, in which the content of the gastrointestinal tract of Tibetan sheep were aseptically collected and isolated through MRS agar medium. The strains were identified by the methods of bacterial morphology, physiological and biochemical characteristics and 16S rDNA gene sequence homology analysis. The probiotic characteristics of the isolated strains were identified by antibacterial test,tolerance test, growth curve acid production ability test and drug sensitivity test. The results showed that the isolated strain could ferment cellobiose, maltose, sucrose and lactose. The 16S rDNA sequence had more than 99% homology with the reference Lactobacillus Sark. The isolates had certain inhibitory effects on Escherichia coli, Salmonella, Staphylococcus aureus, Clostridium perfringens and other strains, which had strong tolerance to high temperature, strong resistance to acidic and alkaline environment, strong acid production ability, and it was sensitive to 11 drugs such as neomycin. The experiment indicates that the Lactobacillus saq obtained in the experiment has strong resistance to acid, alkali and high temperature environment, strong bacteriostatic ability, strong growth rate and acid production ability, which can provide a good reference for the preparation of Tibetan sheep probiotics.
