摘要
目的探讨寻常型银屑病患者外周血微小RNA(miRNA)-155水平变化及其与辅助性T细胞(Th)1/Th2平衡的关系。方法选取95例寻常型银屑病患者为PV组,根据银屑病严重程度分为重度组(n=23)、中度组(n=34)、轻度组(n=38),另选取同期56例体检健康者为对照组。采用qRT-PCR测定外周血miR-155、T-bet、GATA结合蛋白3(GATA3),采用流式细胞术测算外周血Th1、Th2百分比,采用ELISA测定血清干扰素-γ(INF-γ)、IL-2、IL-4、IL-10。采用Pearson相关性分析寻常型银屑病患者外周血miR-155水平与Th1/Th2、T-bet、GATA3的相关性。结果PV组外周血miR-155、Th1百分比、Th1/Th2、T-bet和血清IFN-γ、IL-2水平高于对照组,外周血Th2百分比、GATA3和血清IL-4、IL-10水平低于对照组(P均<0.05)。轻度组、中度组、重度组外周血miR-155、Th1百分比、Th1/Th2、T-bet和血清IFN-γ、IL-2水平逐步升高,外周血Th2百分比、GATA3和血清IL-4、IL-10水平逐步降低(P均<0.05)。Pearson相关性分析显示,寻常型银屑病患者外周血miR-155水平与Th1/Th2、T-bet呈正相关(r分别为0.602、0.749,P均<0.05),与GATA3呈负相关(r=-0.585,P<0.05)。结论寻常型银屑病患者外周血miR-155水平升高,其可能通过调节Th1/Th2平衡介导寻常型银屑病免疫应答,参与寻常型银屑病发生发展。
Objective To investigate the changes of microRNA(miRNA)-155 in the peripheral blood and its relationship with T helper cell(Th)Th1/Th2 balance in patients with psoriasis vulgaris.Methods Ninety-five patients with psoriasis vulgaris were selected as the PV group,and they were divided into the severe group(n=23),moderate group(n=34),and mild group(n=38)according to the severity of psoriasis,and another 56 healthy people with physical examination were selected as the control group during the same period.The miR-155,T-bet,and GATA-binding protein 3(GATA3)levels in the peripheral blood were measured by qRT-PCR,Th1 and Th2 percentages were measured by flow cytometry,and serum interferon-γ(INF-γ),interleukin-2(IL-2),IL-4,and IL-10 levels were measured by ELISA.Pearson correlation was used to analyze the correlation between peripheral blood miR-155 level and Th1/Th2,T-bet and GATA3 in patients with psoriasis vulgaris.Results The miR-155,Th1 percentage,Th1/Th2,T-bet/β-actin levels and serum IFN-γand IL-2 levels in the peripheral bloodwere significantly higher in the PV group than in the control group,while Th2 percentage,GATA3 levels and serum IL-4 and IL-10 levels were significantly lower than those in the control group(all P<0.05).The miR-155,Th1 percentage,Th1/Th2,T-bet levels and serum IFN-γand IL-2 levels in the peripheral blood gradually increased and Th2 percentage,GATA3/β-actin levels and serum IL-4 and IL-10 levels gradually decreased in the mild,moderate and severe groups(all P<0.05).Pearson correlation analysis showed that peripheral blood miR-155 level in the psoriasis vulgaris patients was positively correlated with Th1/Th2 and T-bet(r=0.602,0.749,both P<0.05),and was negatively correlated with GATA3(r=-0.585,P<0.05).Conclusion The level of miR-155 in the peripheral blood of patients with psoriasis vulgaris increases,which may mediate the immune response of psoriasis vulgaris by regulating Th1/Th2 balance and participate in the development and progression of psoriasis vulgaris.
