摘要
目的探究血清微小RNA-155(miR-155)在尿毒症患者血液透析(HD)前后的表达水平及其与辅助性T细胞(Th17)/调节性T细胞(Treg)细胞平衡的关系。方法选取2017年9月至2019年12月本院收治的86例尿毒症患者的临床资料进行研究,将患者按照HD前后分为HD前组和HD后组,并以同期本院94例体检健康者为健康组。比较分析健康组和HD前组的一般临床资料;以实时荧光定量PCR(qRT-PCR)法检测血清miR-155水平;利用流式细胞仪检测Th17、Treg水平,并计算Th17/Treg;检测各组白细胞介素-17(IL-17)、白细胞介素-10(IL-10)水平;分析尿毒症患者HD前后的血清miR-155、Th17、Treg、Th17/Treg与IL-17、IL-10水平的关系,以及血清miR-155表达水平与Th17/Treg细胞比例的相关性。结果HD前组、HD后组的血清miR-155水平、Th17细胞百分比、Th17/Treg细胞比例及IL-17水平均明显高于健康组(均P<0.05);HD前组的miR-155水平、Th17细胞百分比、Th17/Treg细胞比例及IL-17水平均明显高于HD后组(均P<0.05);HD前组、HD后组的Treg细胞百分比及IL-10水平均明显低于健康组(均P<0.05),且HD前组的Treg细胞百分比、IL-10水平均明显低于HD后组(均P<0.05);HD前、HD后尿毒症患者的血清miR-155、Th17、Th17/Treg与IL-17,Treg与IL-10及miR-155表达水平与Th17/Treg细胞比例均呈正相关(均P<0.05),miR-155、Th17/Treg均与IL-10呈负相关(均P<0.05)。结论尿毒症患者的血清miR-155、Th17/Treg细胞比例均明显升高,经HD后,血清miR-155水平、Th17/Treg细胞比例明显降低,两者具有较强相关性,可能通过相互影响,共同在尿毒症发生发展中发挥重要作用。
Objective To investigate the expression of microRNA-155(miR-155)in the serum of uremic patients before and after hemodialysis(HD)and its relationship with the balance of helper T cells(Th17)/regulatory T cells(Treg).Methods Clinical data of a total of 86 patients with uremia who were diagnosed and treated in our hospital from September 2017 to December 2019 were studied,and the patients were divided into pre-HD and post-HD according to before and after HD,and 94 healthy persons in the same period as healthy group.The general clinical data of healthy group and before HD group were compared and analyzed.Level of serum miR-155 was detected by real-time fluorescence quantitative PCR(qRT-PCR);the levels of Th17 and Treg were measured by flow cytometry,and Th17/Treg was calculated;the levels of interleukin-17(IL-17)and interleukin-10(IL-10)in each group were detected;and the correlations between the expression levels of serum miR-155,Th17,Treg,Th17/Treg and IL-17,IL-10,serum miR-155 and the proportion of Th17/Treg cells in uremic patients before and after HD were analyzed.Results The levels of miR-155,the percentage of Th17 cells,the ratio of Th17/Treg cells and the level of IL-17 in uremia patients before and after HD were significantly higher than those in healthy group(all P<0.05).The miR-155 level,Th17 cell percentage,Th17/Treg cell ratio and IL-17 level in pre-HD group were significantly higher than those in post-HD group(all P<0.05).The percentage of Treg cells and the level of IL-10 in uremia patients before and after HD were significantly lower than those in healthy group(P<0.05),and the percentage of Treg cells and the level of IL-10 in uremia patients before and after HD were significantly lower than those in healthy group(P<0.05).Serum miR-155,Th17,Th17/Treg and IL-17 were positively correlated with the proportion of Th17/Treg cells in uremia patients before and after HD(all P<0.05).The miR-155 and Th17/Treg were negatively correlated with IL-10(all P<0.05).Conclusions The serum miR-155 and ratio of Th17/Tre
