雷公藤多苷对湿疹大鼠皮损的预防作用及机制 被引量:6

Preventive effect and mechanism of tripterygium glycosides on skin lesions in eczema model rats

摘要 目的观察雷公藤多苷对湿疹大鼠皮损的预防作用,并探讨其作用机制与组胺受体H1(H1R)/瞬时感受器电位受体1(TRPV1)通路、蛋白酶激活受体2(PAR-2)/TRPV1通路及丝裂原活化蛋白激酶p38(p38 MAPK)通路的关系。方法取健康雄性大鼠50只,随机分为正常组、模型组和雷公藤多苷低、中、高剂量组,每组各10只。正常组正常饲养;模型组和雷公藤多苷低、中、高剂量组采用二硝基氯苯(DNCB)诱导建立湿疹模型,将大鼠腹部及背部脱毛,在腹部皮肤涂抹DNCB-丙酮橄榄油溶液进行致敏,5 d后在背部皮肤涂抹0.5%DNCB-丙酮橄榄油溶液进行激发,在右耳内外侧耳面涂抹1%DNCB-丙酮橄榄油溶液进行激发。于实验第3天开始给药,雷公藤多苷低、中、高剂量组分别给予4.5、9、18 mg/kg雷公藤多苷溶液灌胃,正常组和模型组给予等体积生理盐水灌胃,连续21 d。末次给药24 h后,观察大鼠背部皮肤外观情况;处死大鼠,沿耳基线剪下双耳,用打孔器在左右耳中部取圆形耳片,称取耳片质量,计算耳肿胀度和耳肿胀抑制率;取背部皮肤组织,采用HE染色法进行组织形态理学观察,采用免疫组化法检测组织中H1R、PAR-2、TRPV1蛋白水平,采用Western blotting法检测组织中p38 MAPK、p-p38MAPK蛋白的相对表达量。结果与正常组比较,模型组耳肿胀度增加(P <0.05);与模型组比较,雷公藤多苷低、中、高剂量组耳肿胀度减少(P均<0.05);与雷公藤多苷低剂量组比较,雷公藤多苷中、高剂量组耳肿胀度减少,耳肿胀抑制率增加(P均<0.05)。与正常组比较,模型组背部皮肤出现湿疹病理变化;与模型组比较,雷公藤多苷低、中、高剂量组背部皮肤湿疹病变程度减轻。与正常组比较,模型组皮损组织中H1R、PAR-2、TRPV1、p38 MAPK、p-p38 MAPK蛋白水平升高(P均<0.05);与模型组比较,雷公藤多苷中、高剂量组皮损组织中H1R、PAR-2、TRPV1、p38 MAPK、p-p38 MAPK蛋白水平降低(P� Objective To investigate the preventive effect of tripterygium glycosides on dermatic lesion tissues of eczema rats and to explore the relationships of their mechanism with the signal pathways of histamine H1 receptor(H1R)/transient receptor potential vanilloid(TRPV1) and protease activated receptor 2(PAR-2)/TRPV1,and p38 mitogen-activated protein kinase(p38 MAPK).Methods Totally 50 healthy rats were randomly divided into the normal,model,low-dose,medium-dose and high-dose tripterygium glycosides groups,with 10 rats in each.Eczema rat model was induced by dinitrochlorobenzene(DNCB).The rats’ abdomen and back were depilated,and DNCB acetone olive oil solution was applied to the abdominal skin for sensitization.After 5 days,0.5% DNCB acetone olive oil solution was applied to the dorsal skin for stimulation,and 1% DNCB acetone olive oil solution was applied to the inner and outer ear surface of the right ear for stimulation.We started the administration on the third days of the experiment.The rats in the low-dose,medium-dose and high-dose tripterygium glycosides groups were given 4.5,9,and 18 mg/kg tripterygium glycoside solution by gavage,respectively,and the rats in the normal and model groups were given equal volume normal saline by gavage for 21 days.After the last administration,the appearance of rat dorsal skin was observed;both ears were cut off along the ear baseline,and the round ear pieces were taken from the middle of left and right ears with a punch.The ear pieces were weighed,and the ear swelling and inhibition rate of ear swelling were measured.The dorsal skin tissues were taken.HE staining was used to observe the histopathological characteristics dorsal skin,immunohistochemistry was used to determine the levels of H1R,PAR-2,TRPV1,and Western blotting was used to determine the levels of p38 MAPK and p-p38 MAPK.Results Compared with the normal group,the ear swelling increased in the model group(P < 0.05).Compared with the model group,the ear swelling decreased in the low-dose,medium-dose and high-dose
