三七总皂苷对A549细胞上皮-间充质转化的影响 被引量:5

摘要 目的本研究旨在观察三七总皂苷(Panax notoginseng saponins,PNS)对A549细胞上皮-间充质转化(Epithelial-mesenchymal transition,EMT)的影响及其机制。方法将A549细胞随机分为5组:正常对照组、TGF-β1诱导组(5 ng·mL^(-1),诱导24h)、PNS干预组(低、中、高浓度组分别加入TGF-β15 ng·mL^(-1)和50、100、200μg·mL^(-1)PNS);倒置显微镜下观察各组细胞的形态变化;Elisa法检测细胞上清液中Ⅰ型胶原蛋白(COLⅠ)的表达;Western blot检测各组E-cadherin、Vimentin、磷酸化p38MAPK(P-p38MAPK)蛋白的表达。结果倒置显微镜下TGF-β1诱导的A549细胞由圆形变为长梭形、纺锤形,细胞圆形度降低(P<0.01),PNS干预后细胞圆形度增高(P<0.05);Elisa法检测显示,与正常对照组相比TGF-β1诱导组细胞上清液COLⅠ表达增高(P<0.05),与TGF-β1诱导组比较PNS干预后COLⅠ表达降低(P<0.05);Western blot检测与与TGF-β1诱导组比较,PNS干预后E-cadherin蛋白表达增加(P<0.05),Vimentin蛋白表达降低(P<0.05),磷酸化的p38丝裂原活化蛋白激酶(P-p38 MAPK)表达降低(P<0.05)。结论三七总皂苷可能通过调控TGF-β1/p38MAPK信号通路可以抑制TGF-β1诱导A549细胞发生EMT现象。 Objective To investigate the effect of Panax Notoginseng Saponins(PNS)on epithelial-Mesenchymal transition(EMT)of A549 cells and its mechanism.Methods A549 cells were randomly divided into 5 groups:normal control group,TGF-β1 induction group(5 ng·mL^(-1),24 h induction),and PNS intervention group(low,medium and high concentration groups were added with TGF-β15 ng·mL^(-1)and 50,100 and 200μg·mL^(-1)PNS,respectively).Morphological changes of each group were observed under inverted microscope.Elisa method detected the cell that the supernatant fluid typeⅠcollagen(COLⅠ)expression;The expression of E-cadherin,Vimentin and phosphorylated p38MAPK(P-p38MAPK)were detected by Western blot.Results Under inverted microscope,TGF-β1 induced A549 cells changed from round to long spindle and spindle shape,and the cell roundness decreased(P<0.01).After PNS intervention,the cell roundness increased(P<0.05).Elisa method detection showed that compared with the normal control group,the expression of COLⅠin the cell supernatant of the TGF-β1 induction group increased(P<0.05).Compared with the TGF-β1 induced group after PNS intervention,COLⅠexpressed lower(P<0.05);Compared with TGF-β1 induced group,the expression of E-cadherin protein was increased after PNS intervention(P<0.05),the expression of Vimentin protein was decreased(P<0.05),and the expression of phosphorylated P38 mitogen-activated protein kinase(P-p38 MAPK)was decreased(P<0.05).Conclusion Total saponins of Panax notoginseng may inhibit TGF-β1induced EMT in A549 cells by regulating the TGF-β1/p38MAPK signaling pathway.
