摘要
目的探讨三七总皂苷对人胚肺成纤维细胞MRC-5增殖分化及胶原蛋白合成的作用。方法体外培养人胚肺成纤维细胞MRC-5,随机分为正常对照组、模型组、地塞米松组和三七总皂苷组。采用四氮唑(MTT)比色法检测转化生长因子-β(TGF-β)刺激的成纤维细胞增殖情况;采用免疫细胞化学染色法检测α-平滑肌肌动蛋白(α-SMA)及Ⅰ型、Ⅲ型胶原蛋白表达水平。结果与正常对照组比较,模型组MRC-5细胞增殖比显著升高,α-SMA及Ⅰ型、Ⅲ型胶原蛋白表达水平显著升高;地塞米松组和三七总皂苷组MRC-5增殖比较模型组显著降低,α-SMA和Ⅰ型、Ⅲ型胶原蛋白表达水平较模型组显著降低。结论三七总皂苷可通过抑制MRC-5细胞增殖及分化,抑制人胚肺成纤维细胞Ⅰ型、Ⅲ型胶原蛋白表达,从而延缓及阻止肺纤维化。
Objective To investigate the effect of panax notoginseng saponins(PNS) on proliferation,differentiation and collagen synthesis of human embryonic lung fibroblasts MRC-5.Methods The human embryonic lung fibroblast MRC-5 was cultured in vitro and randomly divided into the normal control group,model group,dexamethasone group,and PNS group.MTT colorimetry was used to detect the proliferation of the transforming growth factor-β(TGF-β)-induced fibroblasts,and immunocytochemical staining was used to detect the expression levels of α-smooth muscle actin(α-SMA) and collagen Ⅰ and collagen Ⅲ.Results Compared with the normal control group,the proliferation ratio of MRC-5 cells in the model group was increased significantly,the expression levels of α-SMA and collagen Ⅰ and collagen Ⅲ in the model group were increased significantly.Compared with the model group,the proliferation ratio of MRC-5 cells in the dexamethasone group and PNS group was decreased significantly,and the expression levels of α-SMA and collagen Ⅰ and collagen Ⅲ in the dexamethasone group and PNS group were decreased significantly.Conclusion PNS can delay and prevent pulmonary fibrosis by inhibiting the proliferation and differentiation of MRC-5 cells and the expression of collagen Ⅰ and collagen Ⅲ in human embryonic lung fibroblasts.
