基于JAK/STAT信号通路观察三七注射液对阿霉素诱导的肾纤维化大鼠炎症因子的影响 被引量:17

摘要 目的:基于JAK/STAT信号通路,观察三七注射液对阿霉素诱导的肾纤维化大鼠炎症因子的影响。方法:144只SD大鼠随机分为4组:正常组(n=36)、假手术组(n=36)、模型组(n=36)和三七注射液组(n=36),每组在给药后第4周、8周、12周三个时间点随机各处死大鼠12只。ELISA法检测血清IL-6、IL-10的含量,Westernblot法进行p-JAK2、p-STAT3蛋白定量分析。结果:(1)模型组、三七组大鼠血清IL-6水平较假手术组均升高(P<0.01),但是,相同时间点三七组IL-6较模型组低(P<0.01)。模型组、三七组IL-10水平较假手术组上升(P<0.05或P<0.01),相同时间点三七组IL-10水平与模型组比较,第4周、第8周较模型组高(P<0.05),但第12周较模型组低(P<0.01)。(2)模型组、三七组大鼠肾组织p-JAK2、p-STAT3蛋白表达水平较假手术组均升高(P<0.01)。与模型组比较,三七组p-JAK2蛋白表达水平均降低(P<0.01);三七组第8周和12周相同时间点p-STAT3蛋白表达较模型组下降(P<0.01)。结论:三七注射液可以调节阿霉素肾纤维化大鼠炎症因子水平,可能与其阻断JAK/STAT信号通路相关,从而发挥抗肾纤维化作用。 Objective:To observe the effect of Sanqi injection on the inflammatory factor in rats model with adriamycin-induced kidney fibrosis based on JAK/STAT signal pathway. Methods:144 rats were divided into 4 groups randomly: normal ]group, sham group, model group and Sanqi injection group. All the 12 rats in each group were Sacrificed at the time point of 4 , 8 , 12 weeks after the treatment. The protein expression of p-JAK2、p-STAT3 were detected through western blot. And the serum level of IL-6、IL-10 was detected by ELISA. Results:(1)The serum of IL-6 in the Sanqi injection group and model group are higher than those in sham group( P <0.01). The serum of IL-6 in the Sanqi injection group is lower than model group at the same time point ( P <0.01). The serum of IL-10 in the Sanqi injection group and model group are higher than those in sham group( P <0.05或 P <0.01). The serum of IL-10 in the Sanqi injection group is higher than model group at the time point of 4, 8 weeks( P <0.05),however,it is lower than model group at 12 weeks.(2)The protein expression of p-JAK2、p-STAT3 in the kidney in the Sanqi injection group and model group are increased, there is statistical significance when compared with those in the sham group. Compared with those in the model group,The protein expression of p-JAK2 in the Sanqi injection group are decreased( P <0.01). The protein expression of p-STAT3 in the Sanqi injection group are decreased at the time point of 8,12 weeks when compared with those in the model group.( P <0.01). Conclusion:Sanqi injection could provide a nephroprotective role by regulating the inflammatory factor. It may be related to blocking the JAK/STAT signaling pathway.
