摘要
目的观察芪明颗粒(QM)对糖尿病视网膜病变(DR)大鼠炎症反应的改善作用,并探讨其可能的作用机制。方法将大鼠随机分为对照组(生理盐水)、DR组(生理盐水)、QM低剂量组(QM 1.2 g·kg^(-1))、QM高剂量组(QM 2.4 g·kg^(-1))和阳性对照组(辛伐他汀40 mg·kg^(-1)),每组10只。酶联免疫吸附(ELISA)法检测血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和C反应蛋白(CRP)水平;HE染色观察视网膜病理学变化;末端脱氧核糖核苷酸转移酶介导的缺口末端标记(TUNEL)法检测视网膜神经节细胞(RGCs)凋亡;Wetern blot法检测视网膜B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、高迁移率族蛋白1(HMGB1)、Toll样受体4(TLR4)和核转录因子κB(NF-κB)蛋白表达。结果治疗后,QM低剂量组、QM高剂量组和阳性对照组血糖水平均较治疗前降低;与对照组比较,DR组、QM低剂量组、QM高剂量组和阳性对照组血糖水平、血清IL-6、TNF-α和CRP水平均升高(P<0.05);与DR组比较,QM低剂量组、QM高剂量组和阳性对照组血糖水平、血清IL-6、TNF-α和CRP水平均降低(P<0.05);与QM低剂量组比较,QM高剂量组和阳性对照组血糖水平、血清IL-6、TNF-α和CRP水平均降低(P<0.05)。HE染色结果显示,对照组视网膜结构完整,RGCs数量较多;DR组视网膜结构破坏,RGCs数量减少;QM各剂量组及阳性对照组视网膜结构较DR组有所改善,RGCs异形细胞减少,其中QM高剂量组和阳性对照组改善效果明显。与对照组比较,DR组、QM低剂量组、QM高剂量组和阳性对照组凋亡指数升高(P<0.05);与DR组比较,QM低剂量组、QM高剂量组和阳性对照组凋亡指数降低(P<0.05);与QM低剂量组比较,QM高剂量组和阳性对照组凋亡指数降低(P<0.05)。与对照组比较,DR组、QM低剂量组、QM高剂量组和阳性对照组视网膜中Bcl-2蛋白和Bcl-2/Bax降低,Bax、HMGB1、TLR4和NF-κB蛋白表达升高(P<0.05);与DR组比较,QM低剂量组、QM高剂量组和阳�
Objective To observe the improvement effect of Qiming Granules(QM)on the inflammatory response in diabetic retinopathy(DR)rats,and explore its possible mechanism.Methods Rats were randomly divided into control group(normal saline),DR group(normal saline),QM low-dose group(1.2 g·kg^(-1) Qiming Granules),QM high-dose group(2.4 g·kg^(-1) Qiming Gra-nules)and positive control group(40 mg·kg^(-1) simvastatin),each with 10 rats.ELISA method was used to detect the serum interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),C-reactive protein(CRP)levels,HE staining was used to observe the retinal pathological changes,TUNEL method was used to detect the retinal ganglion cells(RGCs)apoptosis,and Wetern blot was used to detect the expression of B cell lymphoma-2(Bcl-2),Bcl-2 associated X protein(Bax),high mobility group proteins-1(HMGB1),toll-like receptor 4(TLR4)and nuclear transcription factorκB(NF-κB)protein in the retina.Results After treatment,the blood glucose levels of the QM low-dose group,QM high-dose group and the positive control group were lower than before treatment.Compared with the control group,the blood glucose levels,serum IL-6,TNF-α,CRP increased in DR group,QM low-dose group,QM high-dose group and positive control group(P<0.05).Compared with DR group,the blood glucose levels,serum IL-6,TNF-α,CRP decrease in QM low-dose group,QM high-dose group and positive control group(P<0.05).Compared with the QM low-dose group,the blood glucose level,serum IL-6,TNF-αand CRP decreased in QM high-dose group and positive control group(P<0.05).HE staining showed that the retinal structure of the control group was intact and the number of RGCs was large,the retinal structure of DR group was damaged and the numbers of RGCs were reduced,the retinal structure of the Qiming Granules groups and the positive control group was improved compared with the DR group,and the numbers of RGCs were reduced,the improvement effect of QM high-dose group and positive control group were obvious.Compared with the control group,the apopt
