罗汉果黄素及其主要代谢物抗糖基化活性研究 被引量:2

Study on Antiglycative Activities of Grosvenorine and Its Main Metabolites

摘要 为探讨罗汉果黄素及其主要代谢物的抗糖基化活性,本研究采用体外牛血清白蛋白-果糖-葡萄糖(BSA-Fru-Glu)糖基化体系,探讨罗汉果黄素及其主要代谢产物山奈苷、阿福豆苷和山奈酚对糖基化的抑制作用。结果表明,罗汉果黄素原形物无明显的抗糖基化活性,但其主要代谢物山奈苷、阿福豆苷和山奈酚表现出较强的抗糖基化活性,这些代谢物对荧光AGEs形成的抑制作用的次序为山奈苷<阿福豆苷<山奈酚。1 mmol·L^(-1)山奈酚作用4周后,对AGEs形成的抑制率达到62.6%。进一步研究发现,与模型组BSA-Fru-Glu比较,1 mmol·L^(-1)山奈酚作用4周后,体系的果糖胺含量、蛋白羰基水平和β淀粉样蛋白交联结构分别降低了27.7%、19.4%和49.2%,而蛋白巯基水平上升了16.7%,这些结果表明罗汉果黄素经胃肠道和肠道菌群代谢后,其抗糖基化活性增强。 In order to investigate the in vitro antiglycative activity of grosvenorine and its main metabolites,bovine serum albumin-fructose-glucose(BSA-Fru-Glu)glycosylation system was established to investigate the inhibitory activities of grosvenorine and its main metabolites kaempferitrin,afzelin and kaempferol on glycation.The results showed that the original grosvenorine did not have obvious antiglycative activity,but its main metabolites kaempferitrin,afzelin and kaempferol exhibited strong antiglycative activity.The rank of the inhibitory abilities of these metabolites on formation of fluorescent AGEs was kaempferitrin<afzelin<kaempferol.In addition,compared to BSA-Fru-Glu group,the treatment of 1 mmol·L^(-1) kaempferol for 4 week led to a decrease of the inhibition rate of AGEs formation by 62.6%,as well as reduction of fructosamine content,protein carbonyl and amyloidβcross-linked structure by 27.7%,19.4%and 49.2%,respectively,while an increase of protein thiols by 16.7%.These results suggest that the antiglycative activity of grosvenorine is enhanced after metabolism by gastrointestinal tract and intestinal flora.
