摘要
目的探究槲皮素(QU)通过调控WEE1蛋白激酶(WEE1)在宫颈癌(CC)细胞生长中的作用。方法将Hela细胞实验分为4组:对照组,实验组、实验组+NC组和实验组+WEE1组。对照组正常培养,实验组用50μmol·L^(-1)的QU处理,实验组+NC组先转染空载质粒,再采用50μmol·L^(-1)的QU处理,实验组+WEE1组先转染WEE1过表达质粒,再采用50μmol·L^(-1)的QU处理。用实时荧光定量聚合酶链式反应检测WEE1 mRNA的表达;用细胞计数试剂盒8和克隆形成实验检测细胞增殖,用流式细胞术检测细胞凋亡,用免疫蛋白印迹法检测增殖细胞核抗原(PCNA)和B细胞淋巴瘤2(Bcl-2)蛋白的表达。结果对照组,实验组,实验组+NC组和实验组+WEE1组的WEE1 mRNA的表达分别为1.00±0.14,0.34±0.05,0.37±0.08和0.92±0.12;这4组的细胞活力分别为0.68±0.15,0.45±0.11,0.48±0.15和0.91±0.18;这4组细胞凋亡率分别为(7.82±0.91)%,(27.76±0.80)%,(27.42±1.29)%和(15.36±1.41)%;这4组PCNA蛋白表达分别为1.00±0.11,0.28±0.04,0.32±0.04和1.12±0.15;这4组Bcl-2蛋白表达分别为1.00±0.13,0.32±0.04,0.34±0.04和1.04±0.12。上述指标,对照组与实验组比较,差异均有统计学意义(均P<0.05);实验组和实验组+WEE1组比较,差异均有统计学意义(均P<0.05)。结论 QU通过调控WEE1表达抑制CC细胞的生长。
Objective To explore the role of quercetin (QU) in the growth of cervical carcinoma (CC) cells by regulating WEE1 protein kinase (WEE1).Methods Hela cells were divided into 4 groups:control group,experimental group,experimental+NC group,and experimental+WEE1 group.The control group was cultured normally;the experimental group was treated with 50μm QU;the experimental+NC group was transfected with no-load plasmids and then treated with50μmol·L^(-1)QU;the experimental+WEE1 group was transfected with WEE1 overexpression plasmids and then treated with 50μmol·L^(-1)QU.Quantitative real-time PCR was used to detect the WEE1 m RNA.Cell counting kit-8 and clone formation experiment was used to detect cell proliferation,flow cytometry was used to detect cell apoptosis,respectively.Western blot was used to detect the protein expression of proliferating cell nuclear antigen (PCNA) and B-cell lymphoma-2(Bcl-2).Results WEE1 m RNA expression in control group,experimental group,experimental+NC group and experimental+WEE1 group were 1.00±0.14,0.34±0.05,0.37±0.08 and 0.92±0.12,respectively;the cell viability in the four groups were 0.68±0.15,0.45±0.11,0.48±0.15 and 0.91±0.18,respectively;the cell apoptosis rate in the four groups were (7.82±0.91)%,(27.76±0.80)%,(27.42±1.29)%and (15.36±1.41)%,respectively;the PCNA protein expression in the four groups were 1.00±0.11,0.28±0.04,0.32±0.04 and1.12±0.15,respectively;the Bcl-2 expression in the four groups were 1.00±0.13,0.32±0.04,0.34±0.04 and1.04±0.12,respectively.For the above indicators,the differences between the control group and the experimental group were statistically significant (all P<0.05);the differences between the experimental group and the experimental+WEE1 group were statistically significant (all P<0.05).Conclusion QU inhibits the growth of CC cells by regulating the expression of WEE1.
