摘要
为了探究斑马鱼有氧运动能力的评估方法,为模式动物斑马鱼在运动医学、环境监测、遗传、药理等领域的应用研究奠定基础,本研究利用泳道呼吸仪设计测试方案,测定不同年龄野生型斑马鱼的最大耗氧量和相对临界游泳速度,同时利用微量血乳酸测试仪检测递增游泳速度下斑马鱼的血乳酸水平。结果显示:3月龄、8月龄、14月龄斑马鱼的最大耗氧量和相对临界游泳速度随着年龄的增长而降低;3月龄、8月龄、14月龄斑马鱼的血乳酸阈分别是(2.98±0.86)mmol/L、(2.47±0.29)mmol/L和(3.83±0.25)mmol/L,且分别在其临界游泳速度的60%、80%、80%处出现。本研究建立的斑马鱼最大耗氧量和血乳酸阈检测技术,可作为一种精确评估斑马鱼有氧运动能力的方法。
In order to explore the evaluation method of aerobic exercise capacity in zebrafish and lay a foundation for the research of zebrafish in the fields of sports medicine,environmental monitoring,genetics,pharmacology and so on,a swimming respirometer was used.The maximum oxygen consumption and relative critical swimming speed of wildtype zebrafish of different ages were determined based on the detection scheme designed.Meanwhile,a microblood lactic acid tester was used to detect the lactic acid level in zebrafish blood under progressively increasing swimming speed.The results showed that the maximum oxygen consumption and relative critical swimming speed of 3-,8-and 14-month-old zebrafish decreased with age,and the blood lactate thresholds of 3-,8-and 14-month-old zebrafish were(2.98±0.86)mmol/L,(2.47±0.29)mmol/L and(3.83±0.25)mmol/L,respectively,which appeared at 60%,80%and 80%of the critical swimming speed,respectively.The measurement techniques of maximum oxygen consumption and blood lactate threshold of zebrafish established in this study can be used to accurately evaluate the aerobic exercise capacity in zebrafish.
