玉米赤霉烯酮诱导睾丸细胞线粒体路径的凋亡损伤小鼠生殖能力 被引量:8

摘要 玉米赤霉烯酮(ZEA)污染谷物饲料危害动物健康,研究表明,ZEA能诱导小鼠睾丸细胞的凋亡,并对生殖能力造成影响,但相关分子机制尚未完全探明。本试验将20只雄性小鼠随机分为对照组(CON组)和玉米赤霉烯酮处理组(ZEA组)(20 mg/kg,每天1次,灌服)处理28 d后,观察睾丸病理变化,检测精子活力,评估精子染色质状态和线粒体功能,检测Bax、Bcl-2、Cytc、P53、Caspase-3、ATG5、LC3-Ⅱ、P62、Beclin1的蛋白和mRNA表达。结果显示,与CON组相比,ZEA组中能动精子百分比下降(P<0.01),未成熟精子比例增高、活跃线粒体的活精子比例和精子浓度呈下降趋势,但差异不显著(P>0.05);睾丸生精管畸形、管内生精细胞排列紊乱;Bax、Cytc、P53的mRNA表达上升,其中Bax的mRNA的表达上升差异极显著(P<0.01);Bax、Bcl-2、P53的蛋白表达呈下降趋势,而Caspase-3、Cytc的蛋白表达上升,其中Cytc表达差异极显著(P<0.01);ATG5,LC3-Ⅱ、P62、Beclin1等自噬相关蛋白及mRNA表达上升,但差异均不显著(P>0.05)。结果表明,ZEA通过诱导睾丸组织细胞线粒体路径的凋亡,损伤小鼠生殖能力。 Zearalenone(ZEA) can pollute cereal feed and do harm to animal health.Previous studies have shown that ZEA can induce apoptosis of mouse testicular cells and affect reproductive ability, but the related molecular mechanism has not been fully explored.In this experiment, 20 male mice were randomly divided into control group(CON group) and ZEA group.After 28 days of feed, the pathological changes of testis were observed, the sperm motility was detected, the sperm chromatin status and mitochondrial function were evaluated, and the protein and mRNA expression of Bax, Bcl-2,Cytc, p53,Caspase-3,ATG5,LC3-Ⅱ,p62,Beclin1 were detected.The results showed that compared with CON group, the percentage of motile sperm in ZEA group decreased, the proportion of immature sperm increased, the proportion of active mitochondria and sperm concentration decreased, but there was no significant difference(P>0.05).Pathological examination reveals testicular seminiferous duct malformation, disordered arrangement of spermatogenic cells in testicular duct.The mRNA expression of Bax, Cytc and p53 increased, and the mRNA expression of Bax increased significantly(P<0.05).The protein expression of Bax, Bcl-2 and p53 decreased, while the protein expression of Caspase-3 and Cytc increased, in which the expression of Cytc was significantly different, and the expression of autophagy-related proteins such as ATG5,LC3-Ⅱ,p62 and Beclin1 increased,but the difference was not significant.The results showed that ZEA impaired the reproductive ability of mice by inducing apoptosis of mitochondrial pathway in testicular cells.
