摘要
试验旨在探究玉米赤霉烯酮(zearalenone, ZEA)对妊娠小鼠的毒性损伤及3种中药的缓解作用。将60只8周龄昆明孕鼠随机分为5组,每组12只(设3个重复,每个重复4只)。A组为空白对照组,孕鼠妊娠1~10 d上、下午各灌胃蒸馏水0.2 mL;B组为ZEA组,孕鼠妊娠1~10 d上午灌胃5 mg/kg ZEA,下午灌胃蒸馏水0.2 mL;C、D、E组分别为番茄红素+ZEA组、菟丝子黄酮+ZEA组、泰山磐石散+ZEA组,1~10 d上午灌胃5 mg/kg ZEA,下午分别灌胃20 mg/kg番茄红素、10 mg/kg菟丝子黄酮、1 g/mL泰山磐石散,每次灌胃量均为0.2 mL。母鼠妊娠第11天剖杀,酶联免疫吸附法(ELISA)测定血清雌二醇(E)、卵泡刺激素(FSH)、黄体生成素(LH)和孕酮(P)水平;取孕鼠子宫,显微镜观察ZEA对子宫形态结构的影响;ELISA检测子宫组织匀浆E、FSH、LH和P的含量;实时荧光定量PCR(RT-qPCR)法测定子宫组织匀浆细胞凋亡相关因子mRNA表达量。结果显示,ZEA导致子宫组织结构损伤、水肿;血清和子宫E、FSH和P水平极显著升高(P<0.01),LH水平极显著降低(P<0.01);子宫Bax、Caspase-3和Caspase-8 mRNA相对表达量显著升高(P<0.05),Bcl-2 mRNA相对表达量极显著降低(P<0.01);3种中药均可缓解ZEA引起上述的变化和损伤。结果表明,ZEA对孕鼠产生严重生殖毒性,番茄红素、菟丝子黄酮、泰山磐石散等中药具有缓解其毒性的作用,其中泰山磐石散效果最好。
The purpose of this study was to explore the protective effect of three kinds of traditional Chinese medicines on reproductive toxicity of zearalenone(ZEA) on pregnant rats.Sixty 8-week-old Kunming pregnant mice were randomly divided into five groups with 12 mice in each group(with three replicates and four mice in each replicate).Group A was the control group.Pregnant rats were given 0.2 mL of distilled water in the morning and afternoon from 1 st to 10 th d of pregnancy.Group B was zearalenone(ZEA) group.Pregnant rats were given 5 mg/kg ZEA in the morning and 0.2 mL in the afternoon.Group C,group D and group E were lycopene+ZEA group, dodder flavonoid+ZEA group and Taishan Panshi powder + ZEA group.The rats were given 5 mg/kg ZEA in the morning and 20 mg/kg BW lycopene, 10 mg/kg BW flavonoid and 1 g/mL Panshi powder in the afternoon, respectively, with 0.2 mL each time.The maternal rats were killed on the 11 th d of pregnancy, and the levels of serum estradiol(E),follicle stimulating hormone(FSH),luteinizing hormone(LH) and progesterone(P) were measured by ELISA.The effect of ZEA on uterine morphology was observed under microscope, and the contents of E,FSH,LH and P in uterine tissue homogenate were detected by ELISA.RT-qPCR was used to detect the expression of apoptosis-related factors mRNA in uterine homogenate cells.The results showed that ZEA induced structural damage and edema of uterine tissue,the levels of E,FSH and P in serum and uterus increased significantly,while the level of LH decreased significantly(P<0.01).The relative expression of Bax,Caspase-3and Caspase-8mRNA in uterus increased significantly,while the relative expression of Bcl-2 mRNA decreased significantly(P <0.01),indicating that all three traditional Chinese medicines could alleviate the above changes and injuries caused by ZEA.The results showed that ZEA had serious reproductive toxicity to pregnant mice,and traditional Chinese medicine such as lycopene,dodder flavonoid and Taishan Panshi powder can alleviate its toxicity,among which Ta
