摘要
应用高效液相色谱法(HPLC)确定不同花期金银花的多种有机酸、木犀草苷、花青素成分含量及不同花期间功效成分与抗氧化活性间关系。本研究采集山东平邑地区的7个花期的金银花样本,在中药制备基础上,通过HPLC的方法,完成了对新绿原酸、绿原酸、隐绿原酸、咖啡酸、木犀草苷、花青素的含量测定,通过自由基清除试验明确不同花期样本的抗氧化活性,并进行Pearson相关性分析。结果表明:新绿原酸、绿原酸、咖啡酸、木犀草苷的峰值含量分别为(12.23±0.95)、(82.54±4.83)、(55.74±3.75)、(2.97±0.87) mg/g,均出现在大白期;隐绿原酸、花青素的峰值含量分别为(3.84±1.20)、(0.63±0.04) mg/g,出现在二白期。大白期的自由基清除能力最高;经Pearson相关分析,金银花的绿原酸含量与DPPH、ABTS清除率间均存在显著正相关,新绿原酸、隐绿原酸、木犀草苷含量与ABTS清除率间均存在显著正相关(r>0,P<0.05);花期与花青素含量、DPPH清除率呈显著负相关(r<0,P<0.05)。本研究初步认为,不同花期所含的有机酸、木犀草苷、花青素含量存在差异,大白期的抗氧化活性更强,DPPH自由基清除能力与绿原酸含量有关,ABTS自由基清除能力与新绿原酸、隐绿原酸、木犀草苷含量水平有关。
High performance liquid chromatography(HPLC) determines the content of various organic acids,luteoloside,anthocyanin and antioxidant activity of different flowering periods of honeysuckle.This study collected honeysuckle samples from 7 flowering periods in Pingyi,Shandong Province.Based on the preparation of traditional Chinese medicine,the content determination of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,caffeic acid,luteoloside and anthocyanin was completed,and the antioxidant activity of different flowering samples was defined through free radical clearance test,and Pearson correlation was analyzed.The results showed that the peak content of neochlorogenic acid,chlorogenic acid,caffeic acid,and luteoloside were(12.23 ±0.95),(82.54±4.83),(55.74±3.75),(2.97±0.87) mg/g,appeared in great white period;the peak content of cryptochlorogenic acid and anthocyanin was(3.84±1.20),(0.63±0.04) mg/g,respectively,appeared in second white phase.Free radical clearance capacity was the highest;Pearson correlation analysis showed that chlorogenic acid content of honeysuckle was significantly positively correlated with DPPH and ABTS clearance rate,while the contents of neocchlorogenic acid,cryptochlorogenic acid and luteoloside were significantly positively correlated with ABTS clearance rate(r>0,P<0.05);florescence was negatively correlated with anthocyanin content and DPPH clearance rate(r<0,P <0.05).This study initially believes that the content of organic acid,luteoloside and anthocyanin varies in different flowering periods,the antioxidant activity is greater in the great white period,DPPH free radical clearance ability is related to chlorogenic acid content,and ABTS free radical clearance ability is related to the content level of neochlorogenic acid,cryptochlorogenic acid and luteoloside.
