摘要
研究以斜带石斑鱼(Epinephelus coioides)为试验对象,将分离自健康斜带石斑鱼肠道的3株芽孢杆菌Bacillus aerius B18、 B. aryabhattai B20和B. licheniformis B25以等比例浓度添加至饲料中,制成含菌量1×107CFU/g的饲料,评价其对石斑鱼生长、免疫及肠道菌群的影响。选取120尾平均体重3.5 g的健康石斑鱼,随机分为2组,每组3个重复,每个重复20尾鱼。试验期28 d。结果显示,与对照组相比,复合芽孢杆菌组石斑鱼的体增重和特定生长率显著提高(P<0.05),饵料系数显著降低(P<0.05),石斑鱼血清中酸性磷酸酶、碱性磷酸酶、超氧化物歧化酶、溶菌酶及补体C3的活性显著提高(P<0.05),石斑鱼脾中免疫基因MHCIa、IgM、MHCIIa和CD8a的表达量显著上调(P<0.05),石斑鱼肠道中有利于改善鱼类肠道健康水平的厚壁菌门(Firmicutes)、罗姆布茨菌属(Romboutsia)和鞘氨醇单胞菌属(Sphingomonas)的丰度显著增加(P<0.05)。研究表明,该复合芽孢杆菌具有促进斜带石斑鱼生长、增强鱼体免疫力和改善肠道健康的作用。
The study used Epinephelus coioides as the experimental object, three strains of Bacillus aerius B18, B.aryabhattai B20 and B. licheniformis B25 isolated from the intestinal tract of healthy grouper were added to the feed in equal proportion to produce a 1×107 CFU/g feed to evaluate the effect of diet on growth, immunity and intestinal microflora. The experiment lasted for 28 d. A total of 120 healthy grouper with an average body weight of 3.5 g were randomly divided into two groups, with three replicates in each group and 20 fish in each replicate. The results showed that compared with control group, body weight gain and specific growth rate of grouper in compound Bacillus group were significantly increased(P<0.05), bait coefficient decreased significantly(P<0.05), the activities of acid phosphatase,alkaline phosphatase, superoxide dismutase, lysozyme and complement C3 in serum of grouper were significantly increased(P<0.05), and the expression levels of immune genes MHCIa, IgM, MHCIIa and CD8 a in grouper spleen were significantly up-regulated(P<0.05). The abundance of Firmicutes, Romboutsia and Sphingomonas, which was beneficial for improving fish intestinal health, was significantly increased in compound Bacillus group. The experiment indicates that the compound Bacillus can promote the growth performance, enhance the immunity, improve the intestinal health of grouper.
