摘要
目的:探讨高效液相色谱法(HPLC)测定海水鱼中嘌呤含量的方法和效果。方法:选择新鲜的海鲈鱼、黄花鱼、鳕鱼3种海水鱼类,建立HPLC检测方案,优化色谱条件后测定样品不同部位的腺嘌呤、鸟嘌呤、次黄嘌呤、黄嘌呤含量。结果:4种嘌呤在0.1~400 mg/L的浓度范围内,具有良好的线性关系,相关系数均≥0.998,检出限在0.047~0.106 mg/L,RSD在0.021%~0.689%,平均回收率在94.289%~102.919%。不同海水鱼中的嘌呤含量差异明显,总嘌呤含量海鲈鱼>鳕鱼>黄花鱼;同一海水鱼不同部位的嘌呤含量差异明显,总体上鱼皮>内脏>鱼肉。结论:HPLC法测定海水鱼中嘌呤含量的精密度高、回收率好,是一种可行的检测方案。
Objective:To investigate the method and effect of high performance liquid chromatography(HPLC)for the determination of purine in marine fish.Methods:three kinds of fresh sea bass,yellow croaker and cod were selected to establish an HPLC detection scheme.After optimizing the chromatographic conditions,the contents of adenine,guanine,hypoxanthine and xanthine in different parts of the sample were determined.Results:The four purines had a good linear relationship in the concentration range of 0.1~400 mg/L,the correlation coefficient were all≥0.998,the detection limit was 0.047~0.106 mg/L,the RSD was 0.021%~0.689%,and the average recovery was 94.289%~102.919%.The total purine content of sea bass>cod>yellow croaker;The purine content in different parts of fish in the same sea water was significantly different,in general,fish skin>viscera>fish meat.Conclusion:HPLC is a feasible method for the determination of purine in marine fish with high precision and good recovery.
