化浊解毒方对溃疡性结肠炎大鼠炎性细胞因子、环氧合酶2及前列腺素E_(2)调控作用的研究 被引量:5

摘要 目的观察化浊解毒方对溃疡性结肠炎大鼠炎性细胞因子、前列腺素E_(2)(PGE_(2))、环氧合酶2(COX-2)的影响,探讨其可能作用机制。方法将50只Wistar大鼠随机分为正常组、模型组、美沙拉嗪组、化浊解毒方高剂量组和化浊解毒方低剂量组,每组10只。除正常组外,其余组均采用三硝基苯磺酸/乙醇法建立溃疡性结肠炎模型。造模成功后,化浊解毒方高、低剂量组分别给予化浊解毒方41.6 g/kg、20.8 g/kg灌胃,美沙拉嗪组给予美沙拉嗪0.3 g/kg灌胃,模型组及正常组给予蒸馏水10 mL/kg灌胃,均连续14 d。观察各组大鼠一般情况,计算疾病活动指数(DAI)评分;采用酶联免疫吸附法检测血清白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、PGE_(2)水平;采用免疫组化法检测结肠组织中COX-2表达情况。结果美沙拉嗪组、化浊解毒方高剂量组、化浊解毒方低剂量组DAI评分及血清IL-6、PGE_(2)水平均明显低于模型组(P均<0.05),血清IL-10水平均明显高于模型组(P均<0.05);美沙拉嗪组、化浊解毒方高剂量组DAI评分及血清IL-6、PGE_(2)水平均明显低于化浊解毒方低剂量组(P均<0.05),血清IL-10水平均明显高于化浊解毒方低剂量组(P均<0.05);化浊解毒方高剂量组各指标水平与美沙拉嗪组比较差异均无统计学意义(P均>0.05)。美沙拉嗪组、化浊解毒方高剂量组、化浊解毒方低剂量组COX-2蛋白表达平均光密度值均明显低于模型组(P均<0.05),美沙拉嗪组、化浊解毒方高剂量组均明显低于化浊解毒方低剂量组(P均<0.05),化浊解毒方高剂量组与美沙拉嗪组比较差异无统计学意义(P>0.05)。结论化浊解毒方能够下调IL-6、COX-2表达,抑制PGE_(2)分泌,同时可上调IL-10表达,进而恢复细胞因子的平衡,抑制溃疡性结肠炎大鼠结肠组织的炎症损伤,促进结肠黏膜修复。 Objective It is to observe the effects of Huazhuo Jiedu Decoction on inflammatory cytokines,prostaglandin E_(2)(PGE_(2))and cyclooxygenase 2(COX-2)in rats with ulcerative colitis,and to explore its possible mechanism of action.Methods Fifty Wistar rats were randomly divided into normal group,model group,mesalazine group,high-dose group of Huazhuo Jiedu Decoction and low-dose group of Huazhuo Jiedu Decoction,with 10 rats in each group.Except for the normal group,the rats in the other groups were used to establish ulcerative colitis models by trinitrobenzene sulfonic acid/ethanol method.After successful modeling,the high and low dose groups of Huazhuo Jiedu Decoction were given 41.6 g/kg and 20.8 g/kg of Huazhuo Jiedu Decoction by gavage,respectively,and the mesalazine group was given mesalazine 0.3 g/kg by gavage,the model group and the normal group was given distilled water 10 mL/kg by gavage,all were continuously treated for 14 days.The general conditions of rats in each group were observed,the disease activity index(DAI)was calculated;the levels of serum interleukin-6(IL-6),interleukin-10(IL-10),and PGE_(2)were determined by enzyme-linked immunosorbent assay,the expression of COX-2 in colon tissue was detected by immunohistochemistry.Results The DAI score and serum IL-6 and PGE_(2)levels of the mesalazine group,the high-dose group of Huazhuo Jiedu Decoction,and the low-dose group of Huazhuo Jiedu Decoction were significantly lower than those of the model group(all P<0.05),and the serum IL-10 levels were significant higher than those of the model group(all P<0.05);the DAI score and serum IL-6 and PGE_(2)levels of the mesalazine group and the high-dose group of Huazhuo Jiedu Decoction were significantly lower than those of the low-dose group of Huazhuo Jiedu Decoction(all P<0.05),the serum IL-10 level was significantly higher than that of the low-dose group of Huazhuo Jiedu Decoction(all P<0.05);there was no significant difference in the levels of indexes between the high-dose group of Huazhuo Jiedu Decoction and
