六神丸对健康人上呼吸道菌群结构和多样性的影响 被引量:1

Effect of Liushen Pill(六神丸)on the Constitution and Diversity of Upper Respiratory Tract Flora in Healthy People

摘要 目的基于16S rRNA高通量测序技术探讨六神丸对健康人上呼吸道菌群微生态的影响。方法选择2020年11月2日至11月20日在北京中医药大学及首都医科大学招募的60名健康受试者为研究对象,随机分为对照组和六神丸组各30例。对照组不给予药物干预;六神丸组给予六神丸含服3粒+口服7粒,每日3次,共7天。对照组于招募后第8天、六神丸组于服药后第8天采集受试者咽拭子样本进行16S rRNA高通量测序检测,分析Alpha多样性、Beta多样性和菌群结构差异。结果与对照组比较,六神丸组Chao1指数和Shannon指数增高(P<0.05);对照组和六神丸组非度量多维标定(NMDS)组间分布具有差异,NMDS1胁强系数=0.1590。厚壁菌门、拟杆菌门、变形菌门、梭杆菌门、放线菌门和髌骨细菌门为两组在门水平的优势菌。与对照组比较,六神丸组疣微菌门、硝化螺旋菌门、芽单胞菌门、蓝藻细菌门、绿弯菌门和酸杆菌门相对丰度增高(P<0.05);变形菌门、埃普西隆杆菌门和拟杆菌门相对丰度降低(P<0.05)。结论六神丸对上呼吸道菌群的多样性、丰度和结构均有调节作用,提示六神丸可能通过改善菌群失调对某些呼吸系统疾病起到预防或治疗作用。 Objective To investigate the effect of Liushen Pill(六神丸)on the microecology of upper respiratory tract flora in healthy people based on 16S rRNA high-throughput sequencing technology.Methods A total of 60 healthy subjects were recruited in Beijing University of Chinese medicine and Capital Medical University from November 2nd to 20th,2020,and were randomly divided into control group and Liushen Pill group with 30 in each group.No medication was given to the control group and the Liushen Pill group was treated with 3 pills(sucked)+7 pills(swallowed)of Liushen Pill for 7 days,three times a day.On the 8th day of inclusion or after administration,throat swab samples were collected from the control group or Liushen Pill group through 16S rRNA high-throughput sequencing,to analyze alpha diversity,beta diversity and flora constitution.Results Compared to those in the control group,the Chao1 index and Shannon index in the Liushen Pill group were higher(P<0.05).The distribution of the non-metric multidimensional scaling(NMDS)was significantly different between the control group and the Liushen Pill group,with NMDS1 stress coefficient=0.1590.Firmicutes,Bacteroidetes,Proteobacteria,Fusobacteria,Actinobacteria and Patescibacteria were the dominant phyla in both groups.Compared to the control group,the relative abundance of Verrucomicrobia,Nitrospirae,Gemmatimonadetes,Cyanobacteria,Chloroflexi and Acidobacteria in the Liushen Pill group significantly increased(P<0.05),and the relative abundance of Proteobacteria,Epsilonbacteraeota and Bacteroidetes significantly decreased(P<0.05).Conclusion Liushen Pillhas effects on the diversity,abundance and constituion of the upper respiratory tract flora in healthy people.It is suggested that Liushen Pill may have potential effect on preventing and treating certain respiratory diseases.
