摘要
为增强固定化微生物在生物处理设施中的丰度与污染消除能力,以具备异养硝化-好氧反硝化功用的Pseudomonas poae NL-4为主要供试菌种,并选用小颗粒的活性碳为主要吸附固定化材料,进行了该菌种的吸附-交联固定化条件优化实验与研究。结果显示,在pH为7,菌液与活性碳颗粒的总质量比为1:10,温度30℃,固定化7 h,制备的PseudomoNas poae NL-4,培养72 h后对TN和NH_(4)^(+)-N的消除能力可分别超过5.449、3.919 mg/g。依此为基础,以戊二醛为交联剂,在0.1%的戊二醛溶液中,保持温度30℃,交联4 h,可将固定化PseudomoNas poae NL-4对TN和NH_(4)^(+)-N的72 h消除能力增加到7.920、5.014 mg/g。
In order to enhance the abundance and pollution elimination ability of immobilized microorganisms in biological treatment facilities,Pseudomonas poae NL-4 with heterotrophic nitrification-aerobic denitrification function was used as the main test strain,and the activity of small particles was selected Carbon is the main adsorption and immobilization material,and the optimization experiments and research on the adsorptioncrosslinking and immobilization conditions of this strain have been carried out.The results showed that at pH 7,the total mass ratio of bacterial liquid to activated carbon particles was 1:10,the temperature was 30°C,and the immobilization for 7 h,the prepared PseudomoNas poae NL-4 was cultured for 72 h to TN and NH_(4)^(+)-N.The elimination ability can exceed 5.449 and 3.919 mg/g,respectively.Based on this,using glutaraldehyde as the cross-linking agent,in 0.1%glutaraldehyde solution,keeping the temperature at 30℃,cross-linking for 4 h,the immobilized PseudomoNas poae NL-4 can be used for TN and NH_(4)^(+)-N.The 72 h elimination ability increased to 7.920 and 5.014 mg/g.
