摘要
环状RNA(circRNA)是真核细胞中广泛表达的内源性非编码RNA(ncRNA)的新型簇。与线性RNA转录物生成不同,circRNA经历“反向剪接”过程,在转录后修饰过程中,无5'端帽子结构及3'端多聚腺苷酸尾连接,而是形成稳定的共价闭合环状结构。circRNA在细胞质中数量丰富,进化保守且相对稳定,表明circRNA比其他类型RNA更适合作为生物标志物。自身免疫性疾病由对抗自身抗原的自身免疫反应引发,这种免疫反应损伤人体组织并严重影响患者生活质量。随着circRNA研究深入,发现circRNA可通过各种形式的表观遗传修饰,参与不同免疫反应及免疫疾病发展。circRNA已被证明在多种免疫疾病发生过程中以组织特异性和发病机制相关方式表达,有望成为自身免疫性疾病诊断的生物标志物及治疗靶点。本文结合国内外最新研究,对circRNA在自身免疫性疾病中的研究进展进行综述。
circular RNA(circRNA)was a novel cluster of endogenous non-coding RNA(ncRNA)widely expressed in eu-karyotic cells.Different from production of linear RNA transcripts,circRNA experienced a"reverse splicing"process.In post-tran-scriptional modification process,there was no 5'-end cap structure and 3'-end polyadenylic acid tail,but a stable covalent closed ring structure was formed.circRNA was abundant in cytoplasm,evolutionarily conserved and relatively stable,which indicated that cir-cRNA was more suitable as a biomarker than other types of RNA.Autoimmune diseases were caused by an autoimmune response to self-antigens,which could damage human tissues and seriously affect quality of life of patients.With deepening of circRNA research,it has been found that circRNA enrich various forms of epigenetic modification,participate in development and progression of different immune responses and immune diseases.circRNA has been shown to be expressed in a tissue-specific and pathogenesis-related way during occurrence of a variety of immune diseases,which expected to be a biomarker for diagnosis and therapeutic target of autoim-mune diseases.This article reviewed research progress of circRNA in autoimmune diseases in combination with latest research at home and abroad.
