摘要
目的探讨前列腺癌患者前列腺特异性抗原(PSA)及前列腺特异性膜抗原(PSMA)蛋白表达水平与Gleason评分间的关联性。方法选取保定市第一中心医院诊治的前列腺癌患者80例,对所有患者进行Gleason评分,采用蛋白质印迹法(Wester blot)测定PSA、PSMA的蛋白表达情况,分析比较患者PSA、PSMA的蛋白表达与Gleason评分的相关性,以及在不同分化程度的前列腺癌患者中表达的差异性。结果前列腺癌患者Gleason评分越高,PSMA蛋白表达水平越高,差异均具有统计学意义(P<0.05)。分化程度差的前列腺癌患者PSMA蛋白表达水平明显高于分化中等或良好的患者,差异均具有统计学意义(P<0.05)。不同Gleason评分和分化程度的前列腺癌患者PSA蛋白表达水平没有明显波动,差异均无统计学意义(P>0.05)。Pearson相关性分析证实,前列腺癌患者PSMA蛋白表达水平与Gleason评分呈现正相关性(r=0.871,P<0.01),PSA蛋白表达水平与Gleason评分无相关性(r=0.227,P>0.05)。结论PSMA蛋白表达水平在分化程度差的前列腺癌患者中显著升高,其与Gleason评分呈明显正相关性,提示其可作为前列腺癌Gleason分级的标记物和对激素疗法不敏感的低分化前列腺癌抗体介导免疫治疗的新靶点。
Objective Objective to investigate the correlation between the expression levels of prostate-specific antigen(PSA)and prostate-specific membrane antigen(PSMA)with Gleason score in patients with prostate cancer.Methods 80 patients with prostate cancer in the Baoding fist central hospital hospital were selected.Gleason score was performed on all patients.Western blot was used to determine the protein expression of PSA and PSMA.The correlation between the protein expression of PSA and PSMA and Gleason score was analyzed and compared,as well as the difference of expression in patients with different degrees of differentiation of prostate cancer.Results The higher Gleason score was,the higher PSMA protein expression level was(P<0.05).The expression level of PSMA protein in poorly differentiated prostate cancer patients was significantly higher than that in moderately or well differentiated prostate cancer patients(P<0.05).The expression level of PSA protein in prostate cancer patients with different Gleason scores and differentiation degrees did not fluctuate significantly(P>0.05).Pearson correlation analysis confirmed that there was a positive correlation between the expression of PSMA protein and Gleason score in prostate cancer patient(r=0.871,P<0.01),and there was no correlation between the expression of PSA protein and Gleason score(r=0.227,P>0.05).Conclusion PSMA protein expression level was significantly increased in poorly differentiated prostate cancer patients,which was positively correlated with Gleason score,suggesting that PSMA can be used as a marker of Gleason grade of prostate cancer and a new target of antibody mediated immunotherapy for poorly differentiated prostate cancer which is not sensitive to hormone therapy.
