老鸦糊乙酸乙酯部位化学成分及其抗炎活性 被引量:5

Chemical constituents from ethyl acetate fraction of Callicarpa giraldii and their anti-inflammatory activities

摘要 目的研究老鸦糊Callicarpa giraldii Hesse ex Rehd.乙酸乙酯部位的化学成分及其抗炎活性。方法老鸦糊甲醇提取物乙酸乙酯部位采用硅胶、MCI、Sephadex LH-20、半制备HPLC、重结晶等进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用Griess法测定各化合物对LPS诱导的RAW 264.7细胞模型NO释放量的影响评价其抗炎活性。结果从中分离得到13个化合物,分别鉴定为羽扇豆酮(1)、2α-羟基齐墩果酸(2)、2α-羟基乌苏酸(3)、3β-hydroxy-olean-12-en(4)、邻苯二甲酸二-(2-乙基)己酯(5)、齐墩果酸(6)、4′-methoxy cinnamic acid(7)、7-oxo-15-hydroxydehydroabie tic acid(8)、邻苯二甲酸异二丁酯(9)、(Z)-9-十八碳烯酸甲酯(10)、2α,3β,19α,23-四羟基-齐墩果酸(11)、烟酸(12)、calliphyllin(13)。化合物6、8分别在25、50μmol/L,50、100μmol/L的给药浓度下均具有一定的抗炎活性。结论所有化合物均为首次从该植物中分离得到。化合物6、8具有较好的抗炎活性。 AIM To study the chemical constituents from Callicarpa giraldii Hesse ex Rehd.and their anti-inflammatory activities.METHODS The ethyl acetate fraction of methanol extract from C. giraldii was isolated and purified by silica, MCI, ODS, Sephadex LH-20, semi-preparative HPLC and recrystallization, then the structures of obtained compounds were identified by physicochemical properties and spectral data. Griess method was used to determine the effect of the isolated compounds on LPS induced RAW 264.7 cell model to evaluate their anti-inflammatory activities.RESULTS Thirteen compounds were isolated and identified as lupinone(1), 2α-hydroxyoleanolic acid(2), 2α-hydroxyursolic acid(3), 3β-hydroxy-olean-12-en(4), bis-(2-ethyl)hexyl phthalate(5), oleanolic acid(6), 4′-methoxy cinnamic acid(7), 7-oxo-15-hydroxydehydroabie tic acid(8), isobutyl phthalate(9),(Z)-9-octadecenoic acid methyl ester(10), 2α, 3β, 19α, 23-tetrahydroxy-oleanolic acid(11), niacin(12), calliphyllin(13). Compounds 6, 8 had certain anti-inflammatory activities at 25, 50 μmol/L and 50, 100 μmol/L.CONCLUSION All the compounds are isolated from this plant for the first time. Compounds 6,8 have certain anti-inflammatory activities.
