力学刺激对骨髓间充质干细胞体外分化的影响

Effects of mechanical stimulation on differentiation of bone marrow mesenchymal stem cells in vitro

摘要 目的观察不同强度和时间压应力对骨髓间充质干细胞内成骨细胞、软骨细胞、脂肪细胞相关基因的影响。方法4周龄雄性C57BL/6小鼠处死后,从两侧股骨、胫骨常规方法提取骨髓间充质干细胞,在前期3D水凝胶细胞模型的基础上,次日对细胞进行压应力干预,固定频率为0.5 Hz,选用不同强度、时间梯度,G0为不施加压应力刺激的对照组,其他压应力干预组分别为G1组(1%,4 h)、G2组(1%,8 h)、G3组(1%,12 h)、G4组(2%,4 h)、G5组(2%,8 h)、G6组(2%,12 h)、G7组(3%,4 h)、G8组(3%,8 h)、G9组(3%,12 h),对细胞中的成骨、成脂、软骨基因进行定量检测。结果G1组的成骨相关转录因子抗体(osterix,OSX)表达水平显著高于G2组、G3组、G4组、G7组(P<0.01);G1组的碱性磷酸酶(alkaline phosphatase,ALP)表达水平显著高于G5组、G8组(P<0.01)和G9组(P<0.05);G1组的转录激活因子4(AMP-dependent transcription factor,ATF4)表达水平显著高于G4组、G7组(P<0.01);G1组的增强子结合蛋白(CCAAT/enhancer-binding protein,CEBP)和脂肪细胞基因过氧化物酶体增殖物激活受体γ(peroxisome proliferator-activated receptorγ,PPARγ)显著低于G9组(P<0.01);G1组的软骨细胞基因基质金属蛋白酶13(matrix metalloprotein 13,MMP13)表达水平显著低于G3组(P<0.05)和G5组(P<0.01),并显著低于G8组(P<0.01)。结论1%强度、0.5 Hz频率、持续4 h的压应力可以促进骨髓间充质干细胞的骨向分化。 Objective To research the effects of compressive stress on osteoblasts,chondrocytes,and adipocyte-related genes in bone marrow mesenchymal stem cells under different power and time conditions.Methods Bone marrow mesenchymal stem cells were extracted from femurs and tibias of 4-week old male C57BL/6 mice using conventional methods.The cells were subjected to compressive stress intervention at a frequency of 0.5 Hz.Different intensities and durations were applied for compressive stress intervention on 3D hydrogel cell model,which had been established before.G0 was the control group without compressive stress stimulation,and the other compressive stress intervention groups were G1 group(1%,4 h),G2 group(1%,8 h),G3 group(1%,12 h),G4 group(2%,4 h),G5 group(2%,8 h),G6 group(2%,12 h),G7 group(3%,4 h),G8 group(3%,8 h),G9 group(3%,12 h),respectively.Genes of osteoblasts,adipocytes,and chondrocytes were measured by quantitative testing.Results The expression level of osterix(OSX)in G1 group was significantly higher than that in G2,G3,G4,and G7 group(P<0.01).The expression level of alkaline phosphatase(ALP)in G1 group was significantly higher than that in G5,G8 group(P<0.01)and G9 group(P<0.05).The expression level of AMP-dependent transcription factor 4(ATF4)in G1 group was significantly higher than that in G4 and G7 group(P<0.01).In G1 group,CCAAT/enhancer-binding protein(CEBP)and peroxisome proliferator-activated receptorγ(PPARγ)were significantly lower than these in G9 group(P<0.01).The expression level of matrix metalloprotein 13(MMP13)in G1 group was significantly higher than that of G3(P<0.05)and G5 group(P<0.05),and significantly lower than that of G8 group(P<0.01).Conclusion The compressive stress of 1%intensity,0.5 Hz and 4 h can promote the osteogenic differentiation of bone marrow mesenchymal stem cells.
