摘要
以桦褐孔菌紫外线诱变菌株为材料,利用形态学及ITS分子序列分析相结合的方法进行鉴定,通过热水浸提法、超声辅助浸提法对诱变菌株菌丝体多糖、三萜、黄酮及多酚进行提取,并对各成分含量进行对比分析。结果表明,诱变菌株HZ1与原菌株JL01为同源不同种,诱变菌株生长速率较快为2.974 mm·d^(-1),菌丝体干重为0.385 3 g,其多糖粗提含量为(24.802±0.270) mg·g^(-1)、多酚含量为(17.001±0.010) mg·g^(-1),相比于原菌株JL01含量存在着显著差异,增幅为48.32%、16.06%;而黄酮及三萜含量与原菌株相差并不显著。试验结果为桦褐孔菌的生产加工及品种选育提供理论依据,同时为桦褐孔菌资源的研发提供新思路。
Inonotus obliquus UV mutagenic strain was used as the research material, which was identified by morphology and ITS molecular sequence analysis. The polysaccharides, triterpenoids, flavonoids and polyphenols were extracted from the mycelium of the mutant strain by hot water extraction method and ultrasonic assisted extraction method. The results showed that the mutant strain HZ1 was homologous to the original strain JL01, the growth rate of the mutant strain was 2.974 mm·d^(-1), the dry weight of the mycelium was 0.385 3 g, the content of polysaccharide was(24.802 ± 0.270) mg·g^(-1), the content of polyphenol was(17.001 ± 0.010)mg·g^(-1), there was significant difference compared with the original strain, the increase rate was 48.32% and 16.06%, but the content of flavonoids and it triterpenoids had no significant difference. This study provides a theoretical basis for the production and breeding of I. obliquus, and it provides a new way for the research and development of I. obliquus resources.
