摘要
目的研究下调miR-675-5p对子宫内膜癌细胞(HECCL1)侵袭、迁移影响,并初步探讨其作用机理。方法体外培养HECCL 1细胞,分为对照组、miR-675-5p阴性对照(NC)组和miR-675-5p抑制(inhibitor)组。实时荧光定量PCR(qPCR)法检测人子宫内膜上皮细胞(hEEC)(正常组)和各组HECCL 1细胞miR-675-5p表达情况;噻唑蓝(MTT)法检测各组HECCL 1细胞增殖情况;划痕实验和侵袭实验分别检测各组HECCL1细胞迁移和侵袭能力;蛋白印迹分析法检测各组HECCL 1细胞STAT3蛋白磷酸化水平变化情况。结果与正常组相比,对照组miR-675-5p表达水平显著升高(P<0.05);与对照组和miR-675-5p NC组相比,miR-675-5p inhibitor组HECCL 1细胞miR-675-5p表达水平、细胞存活率、细胞迁移率、侵袭数量和STAT3磷酸化水平显著降低(P<0.05),细胞凋亡率显著升高(P<0.05)。结论下调miR-675-5p可能通过调控STAT3表达,抑制人子宫内膜癌细胞的侵袭和迁移行为。
Objective To investigate the effects of down-regulation of miR-675-5 p on the invasion and migration of endometrial carcinoma cells(HECCL 1)and its mechanism.Methods HECCL 1 cells were cultured in vitro and divided into control group,miR-675-5 p negative control(NC)group and miR-675-5 p inhibitor(inhibitor)group.Real-time fluorescence quantitative PCR(qPCR)was used to detect the expressions of miR-675-5 p in HECCL 1 cells and human endometrial epithelial cells(hEEC)(normal group)in each group;methyl thiazolyl tetrazolium(MTT)assay was used to detect the proliferation of HECCL 1 cells;the migration and invasion abilities of HECCL 1 cells were detected by scratch test and invasion test;the phosphorylation of STAT3 protein were detected by Western blot.Results Compared with that in the normal group,the expression level of miR-675-5 p in the control group was significantly higher(P<0.05);compared with those in control group and miR-675-5 p NC group,the miR-675-5 p expression level,cell survival rate,cell migration rate,invasion number,STAT3 phosphorylation level was significantly lower in miR-675-5 p inhibitor group(P<0.05),and the apoptosis rate was significantly higher(P<0.05).Conclusion Down-regulation of miR-675-5 p may inhibit the invasion and migration of human endometrial carcinoma cells by regulating the expression of STAT3.
