肝爽颗粒对肝损伤大鼠的保护作用 被引量:3

摘要 目的探讨肝爽颗粒对肝损伤模型大鼠肝组织的保护作用及其可能存在的作用机制。方法采用随机数字表法将50只SD大鼠均分为5组:对照组、模型组、肝爽颗粒组(1.8 g/kg、3.6 g/kg、7.2 g/kg),采用LPS/D-GalN腹腔注射法建立肝损伤SD大鼠模型,观察各组大鼠肝组织氧化应激与炎性反应情况、肝组织凋亡及其相关蛋白水平变化。结果各组大鼠肝组织氧化应激检测结果显示模型组、肝爽颗粒组超氧化物歧化酶(SOD)[(270.48±24.05)U/mg、(366.57±23.81)U/mg比(542.62±23.88)U/mg]与谷胱甘肽过氧化物酶(GSH-Px)活性[(23.37±2.90)U/mg、(31.36±3.07)U/mg比(82.99±4.27)U/mg]低于对照组,而丙二醛(MDA)[(7.26±0.34)nmol/mg、(5.43±0.36)nmol/mg比(2.57±0.22)nmol/mg]含量高于对照组(P<0.05);肝爽颗粒组SOD与GSHPx活性高于模型组,而MDA含量低于模型组,存在剂量效应(P<0.05)。各组大鼠肝组织炎性反应检测结果显示模型组、肝爽颗粒组肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)含量均高于对照组(P<0.05),肝爽颗粒组TNF-α、IL-1β、IL-6含量均低于模型组且存在剂量效应(P<0.05)。Tunnel染色结果显示模型组(30.95±2.01)%、肝爽颗粒组(22.74±1.98)%肝组织细胞凋亡率明显高于对照组(2.36±0.68)%,肝爽颗粒组肝组织细胞凋亡率明显低于模型组,且存在剂量效应(P<0.05);蛋白质印迹法(Western blotting)检测显示模型组、肝爽颗粒组肝组织中Bcl-2相关X(Bax)、TLR4、MyD88、核因子κB(NF-κB)蛋白水平高于对照组(P<0.05),而B细胞淋巴瘤-2(Bcl-2)蛋白水平低于对照组(P<0.05);肝爽颗粒组肝组织中Bax、TLR4、MyD88、NF-κB蛋白水平低于模型组(P<0.05),而Bcl-2蛋白水平高于模型组,存在剂量效应(P<0.05)。结论肝爽颗粒可以改善LPS/D-GalN诱导的肝损伤,可能与调节肝组织氧化应激、炎性反应有关。 Objective To explore the protective effect of Ganshuang granules on liver tissues in liver injury model rats and its possible action mechanism.Methods Fifty SD rats were averagely divided into 5 groups:control group,model group and Ganshuang granule group(1.8 g/kg,3.6 g/kg,7.2 g/kg).LPS/D-GalN intraperitoneal injection was applied to establish liver injury SD rat models.The changes of liver tissue oxidative stress and inflammatory response,liver tissue apoptosis and changes in levels of the related proteins in each group were observed.Results The oxidative stress test results of liver tissues in each group showed that activities of SOD[(270.48±24.05)U/mg,(366.57±23.81)U/mg vs.(542.62±23.88)U/mg]and GSH-Px[(23.37±2.90)U/mg,(31.36±3.07)U/mg vs.(82.99±4.27)U/mg]in the model group and Ganshuang granule group were lower than those in the control group,while MDA content[(7.26±0.34)nmol/mg,(5.43±0.36)nmol/mg vs.(2.57±0.22)nmol/mg]was higher than that in the control group(P<0.05).The activities of SOD and GSH in Ganshuang granule group were higher than those in the model group,while MDA content was lower than that in the model group.And there was dose effect(P<0.05).The inflammatory response test results of liver tissue in each group showed that contents of TNF-α,IL-1βand IL-6 in the model group and Ganshuang granule group were higher than those in the control group(P<0.05).The contents of TNF-α,IL-1βand IL-6 in Ganshuang granule group were lower than those in the model group.And there was dose effect(P<0.05).Tunnel staining results showed that apoptosis rate of liver tissue in model group(30.95±2.01)%and Ganshuang granule group(22.74±1.98)%was significantly higher than that in the control group(2.36±0.68)%.The apoptosis rate of liver tissue in Ganshuang granule group was significantly lower than that in the model group.And there was dose effect(P<0.05).Western blotting detection showed that levels of Bax,TLR4,MyD88 and NF-κB proteins in liver tissues in model group and Ganshuang granule group were highe
