摘要
[目的]评估雌性大鼠卵巢切除术后早期椎骨和股骨的骨结构和抗压应力变化特点。[方法] 50只12周龄的雌性SD大鼠随机分为两组,每组25只。分别行双侧卵巢切除和假手术处理。于术后1、4、8、12、16周时每组各随机选取5只大鼠,测量体重,行Micro-CT骨计量、压应力模量检测和组织形态观察。[结果]随时间推移,两组动物体重均显著增加(P<0.05)。术后4~16周,去势组的体重均显著大于假手术组(P<0.05)。Micro-CT骨计量检测方面,随时间推移,去势组椎骨和股骨标本的BV/TV、Conn. D和Tb.N显著下降,而Tb.Sp显著增加(P<0.05),但是Tb.Th无显著改变(P>0.05);相比之下,假手术组不同时间点间的BV/TV、Conn. D、Tb.N、Tb.Th和Tb.Sp均无显著变化(P>0.05)。术后4~16周,去势组BV/TV显著低于假手术组(P<0.05)。力学测试方面,术后4~16周,去势组标本的弹性模量均小于假手术组,但相应时间点,两组间差异均无统计学意义(P>0.05)。组织学方面,随时间推移,与假手术组相比,去势组骨小梁数量减少,间隙增宽,骨小梁连续性变差。[结论]相比力学测试,Micro-CT骨计量检测能更早期反映雌激素缺失造成的骨变化。
[Objective] To evaluate the early variations of bone structure and anti-compressive stress characteristics of vertebrae and femurs in female rats after ovariectomy(OVX). [Methods] Fifty 12-week-old female SD rats were randomly divided into two groups, with 25 rats in each group. The aminals underwent bilateral OVX and sham operation respectively. At 1, 4, 8, 12 and 16 weeks after the OVX, 5 rats in each group were randomly selected for body weight, micro-CT bone morphometry, compressive stress modulus assesments and histomorphologic observation. [Results] The body weight of the rats in both groups increased significantly over time(P<0.05), which proved significantly greater in the OVX group than that in the sham operation group at 4-16 weeks postoperatively(P<0.05). In term of micro-CT bone morphometry, the OVX group had the BV/TV, Conn. D and Tb.N of the vertebrae and femur specimens decreased significantly(P<0.05), the Tb.Sp increased significantly(P<0.05), whereas the Tb.Th remained unchange over time(P>0.05). By contrast, the sham group had no significant variations in BV/TV, Conn. D, Tb.N, Tb.Th and Tb.Sp among different time points(P>0.05). From 4 to 16 weeks after operation, the OVX group had significantly lower BV/TV than the sham group(P<0.05). In term of mechanical test, the OVX group had lower elastic modulus of samples than the sham group from 4 to 16 weeks postoperatively, however, no a significant differnce was noted between the two groups at any corresponding time point(P>0.05). In terms of histologic observation, the number of trabecular bones decreased, the gaps widened, and the continuity of trabecular bones became worse in the OVX group over time. [Conclusion] Compared with the mechanical test, the micro-CT bone morphometry does reflect bone changes induced by estrogen deficiency in an earlier stage.
