天麻总酚闪式提取工艺研究及含量测定 被引量:4

Flash Extraction Process of Total Phenol and Determination of Content from Gastrodia elata

摘要 该试验旨在优化天麻中总酚的闪式提取工艺,并与传统煎煮法及文献报道的提取方法得到的总酚含量进行比较。在单因素试验基础上,采用L9(33)正交试验优化闪式提取条件, Folin-酚比色法测定总酚含量。结果显示,天麻总酚闪式提取最佳条件为提取时间2 min、料液比1∶40 (g/mL)、乙醇体积分数20%。在最佳提取条件下得到的天麻总酚含量为35.25 mg/GAE g。闪式提取法能满足天麻总酚迅速提取的要求。与传统及文献报道的提取方法相比较,闪式提取得到的天麻总酚含量比煎煮法提高了129.40%,而且提取时间大大缩减,该方法提取总酚效率高且工艺简单可行。 To optimize the flash extraction conditions of total phenol from Gastrodia elata(G.elata),and to compared the content of total phenol with the traditional methods and the literatures,on the basis of single factor test,the L9(33) orthogonal test was used to optimize the flash extraction conditions,and the total phenol content was determined by Folin-phenol colorimetric method.The results showed that the optimum flash-extraction conditions for extraction total phenol from G.elata were extraction time 2 min,ratio of material to liquid 1∶40(g/mL) and concentration of ethanol 20%.The total phenol content of G.elata extracted under the optimal conditions was 35.25 mg/GAE g.The method of flash extraction could satisfy the requirement of rapid extraction of total phenol from G.elata.Compared with traditional and literature reports,the total phenol content of G.elata obtained by flash extraction was 129.40% higher than that of decoction method,and the extraction time was greatly reduced.This method had high extraction efficiency and simple process.
