摘要
目的研究酸枣仁汤对老年慢性睡眠剥夺模型大鼠核受体PPARγ及其共激活因子PGC-1α表达的影响。方法将50只雄性Wistar大鼠随机分为空白组、模型组、阳性对照组(0.18mg·kg^(-1)·d^(-1))、酸枣仁汤低、高剂量组(6.48 g·kg^(-1)·d^(-1)、12.96 g·kg^(-1)·d^(-1)),除空白组外,其余各组均皮下注射D-半乳糖,于末次给药后进行多平台水环境睡眠剥夺,造模结束后各组开始灌胃给药,连续给药7天。采用自主活动仪检测大鼠光照环境活动时间、黑暗环境活动时间、总活动时间,采用比色法检测大鼠下丘脑ATP含量的变化,采用RealTime-PCR、Western-blot和免疫组化分别检测大鼠下丘脑核受体PPARγ及其共激活因子PGC-1αm RNA和蛋白表达水平。结果与空白组比较,模型组大鼠光照环境活动时间、黑暗环境活动时间、总活动时间均显著增加(P <0.01);与模型组比较,阳性对照组、酸枣仁汤高剂量组光照环境活动时间、黑暗环境活动时间、总活动时间均显著减少(P <0.05,P <0.01);与空白组比较,模型组下丘脑ATP含量显著降低(P <0.01);与模型组比较,阳性对照组、酸枣仁汤高剂量组下丘脑ATP含量显著升高(P <0.05);与空白组比较,模型组核受体PPARγ及其共激活因子PGC-1αm RNA和蛋白表达水平显著降低(P <0.05,P <0.01);与模型组比较,阳性对照组、酸枣仁汤高剂量组核受体PPARγ及其共激活因子PGC-1αm RNA和蛋白表达水平显著升高(P <0.05,P <0.01)。结论酸枣仁汤有可能基于PPARγ及其共激活因子PGC-1α同时调控昼夜节律与能量代谢。
Objective To study the effect of Suanzaoren Decoction on the expression of PPAR γ and PGC-1α in aged rats with chronic sleep deprivation. Methods 50 male Wistar rats were randomly divided into control group, model group, positive control group(0.18 mg·kg^(-1)·d^(-1)), low and high dose Suanzaoren Decoction groups(6.48 g·kg^(-1)·d^(-1), 12.96 mg·kg^(-1)·d^(-1)). Except for the control group, D-galactose was subcutaneously injected into the other groups. After the last administration, the rats were subjected to multi platform water environment sleep deprivation. After the end of modeling,the rats in each group were given intragastric administration for 7 consecutive days. The activity time in light environment, dark environment and total activity time were detected by autonomic activity analyzer. The content of ATP in hypothalamus was detected by colorimetry. The mRNA and protein expression levels of PPAR γ and its coactivator PGC-1 α in hypothalamus were detected by real time PCR, Western blot and immunohistochemistry. Results Compared with the control group, the light environment activity time, dark environment activity time and total activity time of the model group were significantly increased(P<0.01);compared with the model group, the light environment activity time, dark environment activity time and total activity time of the positive control group and the high dose Suanzaoren Decoction group were significantly decreased(P<0.05, P<0.01);compared with the control group, the light environment activity time, dark environment activity time and total activity time of the model group were significantly increased(P<0.05, P<0.01). The content of ATP in hypothalamus of model group was significantly lower than that of model group(P<0.01);the content of ATP in hypothalamus of positive control group and high-dose Suanzaoren Decoction group was significantly higher than that of model group(P<0.05);the content of PPAR γ and PGC-1α in model group was significantly higher than that of the control group(P<0.05, P
