薄皮甜瓜品种翡翠的SSR快速鉴定技术的应用 被引量:1

摘要 为了准确快速建立薄皮甜瓜品种翡翠的SSR鉴定体系,利用234对甜瓜SSR特异性引物对翡翠品种亲本进行引物筛选,筛选出3对扩增出差异片段的引物,结合商品种F1代进行验证,获得1条最优引物。在开展田间人为掺杂母本并定植后,大田和室内2种鉴定方法对同一定植苗开展一一对应鉴定试验,结果发现室内分子鉴定与大田鉴定符合率达100%;再对生产销售近2 a的样本鉴定发现,室内鉴定与大田鉴定符合率达99%以上。结果表明,薄皮甜瓜品种翡翠的SSR快速鉴定技术可以应用于实际生产中。 In order to accurately and quickly establish an SSR identification system for the thin-skinned melon cultivar Feicui,using 234 pairs of melon SSR-specific primers to screen the parents of Feicui,three pairs of primers that amplified the different fragments were screened out,and then combining with the commodity species F1 for verification,one optimal primer was obtained.After the field artificial adulteration of female parents was carried out and planted,the two identification methods of field and indoor carried out one-to-one identification tests on the same planting seedlings and found that thecoincidence rate of indoor molecular identification and field identification was 100%.The appraisal of samples from production and sales for nearly two years found that the coincidence rate of indooridentification and field identification was over 99%.The results showed that the SSR rapididentification technology for thin-skinned melon cultivar Feicui could be applied in actual production.
